目的 研究人巨细胞病毒(HCMV)感染对人胚肺成纤维(HEL)细胞分泌趋化因子:白细胞介素-8(IL-8)及正常T细胞表达和分泌,受活化调节的蛋白(RANTES)的影响及意义.方法 用逆转录-聚合酶链反应(RT-PCR)法检测IL-8 mRNA和RANTES mRNA的表达,用双抗体夹心ELISA法测定IL-8和RANTES蛋白的分泌.结果 HEL细胞感染HCMV后,IL-8无论从mRNA水平或蛋白质水平,均随感染时间的延长,表达逐渐增加,至感染后72 h,IL-8 mRNA约为对照组的12倍,IL-8蛋白约为对照组的9倍.感染后8 h可测出RANTES mRNA,24 h达到高峰,其后虽有所下降,但仍维持较高水平;RANTES蛋白的分泌在感染后24 h达到高峰,随后急剧下降,至72 h基本无表达.结论 HCMV感染HEL细胞后,诱导IL-8基因转录及IL-8蛋白分泌增加,从而趋化中性粒细胞至感染部位,并促进中性粒细胞的移行而传播病毒.HCMV可通过模拟β族趋化因子受体而下调细胞外RANTES蛋白的分泌,逃避感染时的局部免疫应答.

목적 연구인거세포병독(HCMV)감염대인배폐성섬유(HEL)세포분비추화인자:백세포개소-8(IL-8)급정상T세포표체화분비,수활화조절적단백(RANTES)적영향급의의.방법 용역전록-취합매련반응(RT-PCR)법검측IL-8 mRNA화RANTES mRNA적표체,용쌍항체협심ELISA법측정IL-8화RANTES단백적분비.결과 HEL세포감염HCMV후,IL-8무론종mRNA수평혹단백질수평,균수감염시간적연장,표체축점증가,지감염후72 h,IL-8 mRNA약위대조조적12배,IL-8단백약위대조조적9배.감염후8 h가측출RANTES mRNA,24 h체도고봉,기후수유소하강,단잉유지교고수평;RANTES단백적분비재감염후24 h체도고봉,수후급극하강,지72 h기본무표체.결론 HCMV감염HEL세포후,유도IL-8기인전록급IL-8단백분비증가,종이추화중성립세포지감염부위,병촉진중성립세포적이행이전파병독.HCMV가통과모의β족추화인자수체이하조세포외RANTES단백적분비,도피감염시적국부면역응답.
Objective To investigate the effect of human cytomegaloviurs(HCMV)infection on the expression of interleukin-8(IL-8)and regulated on activation,normal T expressed and secreted (RANTES)SO as to explore the possible mechanism of the immunopathologieal changes caused by HCMV infection.Methotis Expression of IL-8 and RANTES mRNA was detected by PT-PCR.Secretion of IL-8 and RANTES protein was quantitated by enzyme linked immunosorbant assay(ELISA).Results After HCMV infection,the expression of IL-8 in HEL cells was increased gradually concomitant with the prolonged infection time at both the transcriptional level and the protein level.By the time of 72 h after infection,as compared with mock-infected cells,IL-8 mRNA expression was increased up to 12-fold and IL-8protein uD to 9-fold in HCMV-infected cells.The expression of RANTES mRNA was detectable at 8 h after infection,with a maximum at 24 h after infection,since then it remained at a relatively high level.The expression of RANTES protein peaked at 24 h after infection,then dropped sharply and was almost undetectable by the time of 72 h after infection.Conclusion HCMV infection of HEL cells may induce the transcription of IL-8 gene as well as the production of IL-8 protein,therefore enhancing HCMV spread via stimulating neutrophil migration.HCMV may down-regulate extra-cellular production of RANTES protein via mimicing the receptor of β family chemokine as a mechanism to evade the local immune response to infection.
