目的 建立玄参药材的毛细管电泳指纹图谱(CEFP).方法 采用毛细管区带电泳法,以50 mmol·L-1硼砂为背景电解质,运行电压12.1 kV,检测波长280 nm,重力进样20 s(高度8.5 cm),以10个不同产地药材的电泳图谱建立了玄参CEFP.用"中药色谱指纹图谱超信息特征数字化评价系统3.0"软件获得相关指纹峰特征参数,用双定性相似度(SF与S′F)和双定量相似度(C与P)分别评价.对玄参CEFP进行了破坏性试验考察并考察了大峰和小峰分别缺失时4个相似度指标变化特征.结果 以哈巴俄苷峰为参照物峰,按共有峰出现率100%确定10个共有指纹峰,建立了玄参CEFP.采用DQDQS法评价出3批质量完全合格,4批含量明显偏低,3批含量明显偏高.破坏性试验结果表明玄参CEFP指纹在碱性条件下极易降解,其次在酸性条件和氧化作用下也发生降解.采用混批勾兑技术按含量相似度105%计算了两类含量高低不同样品的勾兑系数.结论 由SF与S′F和C与P构成的双定性双定量相似度(DQDQS)法能同时监测大峰和小峰的变动和缺失,能准确的解决色谱指纹图谱的宏观定性和定量评价的关键问题,所建立的CEFP为玄参药材质量控制提供了新方法.

Objective To establish the capillary electrophoresis fingerprint (CEFP) of Raidix Scrophulariae. Methods CEFPs were obtained by capillary zone electrophoresis, in which the background electrolyte was 50 mmol·L-1 sodium borate. The voltage was 12.1 kV and the wavelength was set at 280 nm. The fingerprints were produced from electropherogram of 10 batches of samples. The 10 electropherograms were assessed by the software of the Digitized Evaluation System for Super-information Characteristics of the TCM Fingerprints, and also evaluated by dual qualitative similarities (SF and S′F) and dual quantitative similarities (C and P). The investigation of 5 degradation tests was made,and the variation of the 4 indexes was discussed when different large and small peaks were lost.Results Ten co-possessing peaks were selected as the characteristic fingerprint peaks of Raidix Scrophulariae when choosing harpagoside peak as the referential peak.The chemical component quantity of 3 batches of samples was completely qualified, while the dual quantitative similarities of 4 batches were very low and the contents of 3 batches were relatively high. The degradation test indicated that the fingerprints were much more unstable in basic condition and secondary in acidic condi-tion. Taking 105% as a norm,the blending coefficient between the high content crude drugs and the lower ones was successfully calculated.Conclusion SF can clearly reflect the chemical constituent distribution, and C can reflect the total contents of the sample, but both are seriously influenced by large peaks and can not reflect the missing of small peaks. Both S′F and P are of equal weight to the integrated area of all fingerprint peaks, and they can sensitively reflect the missing of small peaks. The dual qualitative similarities (SF and S′F) and dual quantitative similarities (C and P) are combined to accurately achieve the qualitative and quantitative assessment of quality of traditional Chinese medicine,for the changes or lacks of both big peaks and small peaks can be monitored simultaneously for fingerprints evaluation. It certainly can serve as a novel way to identify and control the quality of Raidix Scrophulariae.
