用培养24h的苏云金杆菌,经稀释处理,显微计数,然后将定量的菌液涂布于平板上,用紫外线对4个Bt亚种(Bt-1.897,Bt-1.903,Bt-1.905,Bt-1.959)进行不同照射时间处理后,再于平板上培养24h,结果表明:Bt-1.897在紫外线处理15min仍然还有活性,Bt-1.903在紫外线处理12min基本上全部失活,Bt-1.905和 Bt-1.959这2个菌株在紫外线处理12min时还有活性,但在处理15min后全部失活.说明不同种的苏云金杆菌对紫外线的抗性有一定差异.经紫外线处理后 Bt-1.897、Bt-1.903、Bt-1.905、Bt-1.959各自有部分菌落发生明显的变异,菌落大小明显小于原始菌落,并且发生变异的菌落呈现出新的颜色,经涂片后在油镜下观察发现,其菌体要比原始菌体小,为明显的变异菌株.
用培養24h的囌雲金桿菌,經稀釋處理,顯微計數,然後將定量的菌液塗佈于平闆上,用紫外線對4箇Bt亞種(Bt-1.897,Bt-1.903,Bt-1.905,Bt-1.959)進行不同照射時間處理後,再于平闆上培養24h,結果錶明:Bt-1.897在紫外線處理15min仍然還有活性,Bt-1.903在紫外線處理12min基本上全部失活,Bt-1.905和 Bt-1.959這2箇菌株在紫外線處理12min時還有活性,但在處理15min後全部失活.說明不同種的囌雲金桿菌對紫外線的抗性有一定差異.經紫外線處理後 Bt-1.897、Bt-1.903、Bt-1.905、Bt-1.959各自有部分菌落髮生明顯的變異,菌落大小明顯小于原始菌落,併且髮生變異的菌落呈現齣新的顏色,經塗片後在油鏡下觀察髮現,其菌體要比原始菌體小,為明顯的變異菌株.
용배양24h적소운금간균,경희석처리,현미계수,연후장정량적균액도포우평판상,용자외선대4개Bt아충(Bt-1.897,Bt-1.903,Bt-1.905,Bt-1.959)진행불동조사시간처리후,재우평판상배양24h,결과표명:Bt-1.897재자외선처리15min잉연환유활성,Bt-1.903재자외선처리12min기본상전부실활,Bt-1.905화 Bt-1.959저2개균주재자외선처리12min시환유활성,단재처리15min후전부실활.설명불동충적소운금간균대자외선적항성유일정차이.경자외선처리후 Bt-1.897、Bt-1.903、Bt-1.905、Bt-1.959각자유부분균락발생명현적변이,균락대소명현소우원시균락,병차발생변이적균락정현출신적안색,경도편후재유경하관찰발현,기균체요비원시균체소,위명현적변이균주.
After cultured for 24h,Bacillus thuringiensis was diluted, counted under microscope, and coated on plates. Bt-1.897,Bt-1.903,Bt-1.905,and Bt-1.959 were irradiated with ultraviolet at different times,and then cultured for 24h. The results showed that Bt-1.897 still had activity after ultraviolet irradiation for 15min, and Bt-1.903 was deactivated after ultraviolet irradiated 12min.Bt-1.905 and Bt-1.959 still had activity after ultraviolet irradiation for 12min, but deactivated after they were irradiated 15min. The different subspecies of Bacillus thuringiensis had different resistance to ultraviolet after ultraviolet irradiation, and some of the colonies varied obviously. The size of colonies were obvious smaller than that of the original colonies. New color was observed with an optical microscope under an oil-immersion lens, and the cells of mutant strains were obvious smaller than original cells.
