目的 研究Notch1蛋白在人肺腺癌组织中的表达及意义,探讨Notch1受体在人肺腺癌发生中的作用.方法 前期研究初步筛选出Notch1蛋白在肺腺癌组织中有强表达,本研究收集我院及新华医院2009年1月至2010年5月胸外科肺癌手术并经术后病理切片明确为肺腺癌的标本以及正常肺组织标本,以实时荧光定量PCR及Western blot方法分别检测肺癌组织中Notch1 mRNA及蛋白的表达水平,进一步探讨Notch1在肺腺癌中所发挥的作用.结果 实时荧光定量PCR分析显示,Notch1mRNA的表达在肺腺癌组织及正常肺组织中分别为1.99±1.40和1.50±1.03,组间比较差异有统计学意义(P <0.05);Western blot分析显示,Notch1蛋白在肺腺癌组织及正常肺组织中分别为0.64±0.25和0.14±0.07,组间比较差异有统计学意义(P<0.01).结论 Notch1信号在肺腺癌的发病机制中可能起促进作用.

Objective To investigate the expression of Notch1 and its role in the pathogenesis in adult adenocarcinoma of lung.Methods The high expression of Notch1 was detected in lung adenocarcinoma in our previous study.In this investigation,operation specimens of adenocarcinoma of lung that were proved by pathological sections were collected.Notch1 mRNA and protein in lung adenocarcinoma were measured with real-time fluorescence quantitative PCR and Western blot respectively to investigate the role of Notchl in adenocarcinoma of lung.Results It was shown by real-time fluorescence quantitative PCR that the expression of Notch1 mRNA in lung adenocarcinoma group (1.99+1.40) was higher than that in normal lung group (1.50+1.03) ( P <0.05).The expression of Notch1 protein in lung adenocarcinoma group (0.64+0.25) was significantly higher than that in normal lung group ( 0.14 + 0.07 ) ( P < 0.01 ) by Western blot.Conclusions Notch1 signal may play a stimulative role in the pathogenesis of lung adenocarcinoma.
