目的 优化葎草多糖的超声辅助提取工艺,并研究其抑菌活性.方法 以多糖中的葡萄糖含量为指标,对超声波功率、料液比和超声时间进行L_9(3~4)正交实验,确定超声辅助提取葎草粗多糖的最佳工艺.用DE-52交换柱对葎草粗多糖进行分离,以滤纸片法研究葎草粗多糖和多糖分离组分的抑菌能力.结果 以水为提取溶剂,料液比1:28(g/ml),超声功率120W,超声90 min提取2次,葎草多糖的得率为0.755%.葎草粗多糖的抑菌效果具有一定的剂量依赖性;各多糖组分中HSP-Ⅰ、HSP-Ⅲ、HSP-Ⅳ发挥主要抑菌作用,其中HSP-Ⅰ效果最显著,HSP-Ⅱ抑菌效果不明显,且各组分间表现出协同关系.结论 超声波辅助提取葎草多糖方法可行,得率高.葎草多糖具有抑菌活性.

Objective To optimize the ultrasound-assisted extraction method and to study the antibacterial activity of polysaccharides from Humulus scandens. Methods Taking the content of glucose as observation index, we optimized the extraction process by L_9(34)orthogonal experiment. The Humulus scandens polysaccharides (HSP) was purified into HSP-Ⅰ, HSP-Ⅱ, HSP-Ⅲ and HSP-Ⅳ by ion exchange chromatography with DE-52. The antibacterial activity of each component was tested by means of filter paper.Results The optimal extraction conditions were obtained as follows: water refluxing, a ratio of solid -liquid as 1g:28ml,120W for the intension of ultrasonic power, extracting time 90min and extracting two times. Under aforesaid optimization conditions, the yield of polysaccharides was 0.755%. Antibacterial experiments indicated that the inhibitory effect of crude polysaccharides showed dose-dependent. Among the isolated components, HSP-Ⅰ, HSP-Ⅲ, HSP-Ⅳ played main antibacterial roles and HSP-Ⅱ was ineffective. It was cooperative with different isolated components. Conclusion The yield of Humulus Scandens polysaccharides is high with ultrasound-assisted extraction method. The Humulus scandens polysaccharides has obvious antibacterial action.
