目的 探讨大柴胡汤对大鼠无水乙醇性胃黏膜损伤的防护作用,研究其组方中单味有效药物.方法 66只雄性SD大鼠按随机数字表法分为正常对照组、模型组和9个治疗组(大柴胡汤组、柴胡组、黄芩组、枳实组、半夏组、大黄组、白芍组、生姜组、大枣组),每组各6只.正常对照组和模型组以生理盐水2ml灌胃,各治疗组以相应的药液2ml灌胃,2h后,正常对照组予生理盐水1ml灌胃,其余各组均予无水乙醇1ml灌胃.灌胃2h后,11组大鼠均乙醚麻醉下剖腹取胃,观察胃黏膜改变,计算溃疡指数和溃疡抑制率;光镜下观察病理组织学改变.制备胃组织匀浆,检测超氧化物歧化酶(SOD)和丙二醛(MDA)含量.结果 大体观察及病理组织切片显示,大柴胡汤组、黄芩组、枳实组、生姜组、大枣组胃黏膜与正常对照组接近,未见明显损伤;柴胡组、半夏组、大黄组及白芍组大鼠胃黏膜与模型组相似,胃黏膜有明显损伤.大柴胡汤组、黄芩组、枳实组、生姜组、大枣组与正常对照组SOD、MDA含量比较,差异无统计学意义(P>0.05);而模型组、柴胡组、半夏组、大黄组和白芍组与正常对照组比较,SOD含量降低[(121±13)、(244 ±23)、(252±16)、(242±18)、(249±12) U/g比(357±21) U/g]、MDA含量升高[(15.0±1.6)、(14.4±1.3)、(13.9±1.0)、(14.5±1.1)、(14.1 ±0.7) μmol/g比(9.8±0.6) μmol/g],差异均有统计学意义(均P<0.05).结论 大柴胡汤及其单味药黄芩、枳实、生姜和大枣可有效对抗无水乙醇致大鼠胃黏膜损伤,其防护胃黏膜的机制很可能是通过抗氧化起作用.

목적 탐토대시호탕대대서무수을순성위점막손상적방호작용,연구기조방중단미유효약물.방법 66지웅성SD대서안수궤수자표법분위정상대조조、모형조화9개치료조(대시호탕조、시호조、황금조、지실조、반하조、대황조、백작조、생강조、대조조),매조각6지.정상대조조화모형조이생리염수2ml관위,각치료조이상응적약액2ml관위,2h후,정상대조조여생리염수1ml관위,기여각조균여무수을순1ml관위.관위2h후,11조대서균을미마취하부복취위,관찰위점막개변,계산궤양지수화궤양억제솔;광경하관찰병리조직학개변.제비위조직균장,검측초양화물기화매(SOD)화병이철(MDA)함량.결과 대체관찰급병리조직절편현시,대시호탕조、황금조、지실조、생강조、대조조위점막여정상대조조접근,미견명현손상;시호조、반하조、대황조급백작조대서위점막여모형조상사,위점막유명현손상.대시호탕조、황금조、지실조、생강조、대조조여정상대조조SOD、MDA함량비교,차이무통계학의의(P>0.05);이모형조、시호조、반하조、대황조화백작조여정상대조조비교,SOD함량강저[(121±13)、(244 ±23)、(252±16)、(242±18)、(249±12) U/g비(357±21) U/g]、MDA함량승고[(15.0±1.6)、(14.4±1.3)、(13.9±1.0)、(14.5±1.1)、(14.1 ±0.7) μmol/g비(9.8±0.6) μmol/g],차이균유통계학의의(균P<0.05).결론 대시호탕급기단미약황금、지실、생강화대조가유효대항무수을순치대서위점막손상,기방호위점막적궤제흔가능시통과항양화기작용.
Objective To study the protective effects of daisaikoto and its components on alcohol-induced gastric mucosal injury and to explore the mechanism.Methods Sixty-six male Sprague Dawley rats were randomly divided into 11 groups with 6 rats in each group,namely control group,model group and 9 therapeutic groups ( groups of daisaikoto,bupleurum,baicalin,zhishi,banxia,rhubarb,paeony,ginger,jujube).The rats in control group and model group were given 2 ml of normal saline intragastrically; rats in other groups were given 2 ml of different solutions of above drugs respectively.Two hours later,rats in control group were given 1 ml of normal saline and other groups were given 1 ml of absolute alcohol intragastrically.After 2 hours,all rats were sacrificed and gastric mucusal injury index and injury inhibition rates were calculated.Pathological changes in gastric mucosa were observed after HE stain.superoxide dismutase (SOD) and Maleic dialdehyde ( MDA)were measured in gastric homogenate.Results The gastric mucosa in control group was intact and the tissue structure was orderly arranged.There are mucous hemorrahge and necrosis in model group.The mucous changes in groups of bupleurum,banxia,rhubarb and paeony were similar to those in model group.The mucosal injury was mild in the groups of daisaikoto,baicalin,zhishi,ginger,jujube,with no significant difference from control group.SOD significantly decreased while MDA increased in model group (P < 0.05 ).Daisaikoto,baicalin,zhishi,ginger,jujube can antagonize the decrease of SOD and the increase of MDA.Conclusions Daisaikoto efficiently antagonizes alcohol-induced gastric mucosal injury.Its components of baicalin,zhishi,ginger and jujube have the similar effects.Daisaikoto and its effective components protect gastric mucosa from alcohol-induced injury possibly through the effects of oxidation resistance.
