目的 探讨白藜芦醇对Jurkat T细胞T-bet/GATA-3表达的影响及其与调节Th1/Th2细胞分化作用机制之间的关系.方法 不同浓度的白藜芦醇刺激Jurkat T细胞48 h后,用ELISA法检测Th1/Th2细胞因子表达谱及用RT-PCR检测T-bet和GATA-3 mRNA表达的变化.结果 白藜芦醇抑制T-bet mRNA和GATA-3 mRNA的表达(F=6308,P<0.05;F=3262,P<0.05).对Jurkat T细胞分泌细胞因子IFN-γ、IL-10的表达均起抑制作用,并具有浓度效应(F=2604,P<0.05;F=2465,P<0.05).结论 白藜芦醇抑制Jurct T细胞由Th0向Th1细胞分化可能是通过降低转录因子T-bet表达水平进行调节,对Th2细胞的抑制作用则可能通过抑制转录因子GATA-3途径进行负调节,并具有浓度效应.

Objective To investigate the role of resveratrol in the gene expression of T-bet/GATA-3 in Jurkat T cells,and to explore the mechanisms underlying this sensitizing effect of the change of TH cell subpopulation group. Methods The related cytokines about Thl/Th2 were detected by ELISA and T-bet and GATA-3 mRNA were detected by RT-PCR after Jurcat T cell are cultured with different doses of Resveratrol for 48 hours. Results Resveratrol inhibited the expressions of mRNA of T-bet and GATA-3 ( F = 6308, P < 0. 05; F = 3262,P< 0.05) and the secretions of the related cytokines about Thl/Th2(F = 2604,P <0. 05; F =2465, P<0. 05). And these two effects are all concentration related. Conclusion Resveratrol could inhibit Th0 cells to differentiate to Thl cells by down-regulating transcription factor T-bet. Resveratrol also could inhibit Th2 cells differentiation from decreasing the expression of mRNA of GATA-3. And these two effects are all concentration related.
