目的探讨微小RNA(miRNA)-221和miRNA-222表达与胶质瘤病理分级及胶质瘤患者预后的关系.方法收集手术切除的胶质瘤标本120例,病理学分级Ⅰ级19例、Ⅱ级31例、Ⅲ级38例、Ⅳ级32例；采用实时聚合酶链反应(Real-time PCR)及荧光原位杂交(FISH)法检测miRNA-221和miRNA-222的表达.对进行标准治疗的胶质瘤患者进行4年随访.结果肿瘤病理级别随miRNA-221和miRNA-222表达的升高而增加,组间比较差异有统计学意义(x2=43.53,P＜0.01)；生存时间随miRNA-221和miRNA-222表达升高而减少,各组间差异有统计学意义(x2 =55.63,P＜0.01).结论 miRNA-221和miRNA-222的表达与胶质瘤的病理学分级及预后密切相关,可以作为独立的预后因素.

Objective To investigate the relationship between the expression of microRNA ( miRNA)-221 and miRNA-222 and pathological grade of glioma and prognosis of patients with glioma.Methods The expression of miRNA-221 and miRNA-222 was detected by real-tiem polymerase chain reaction ( Realtime PCR) and FISH in 120 cases of glioma.Of the 120 cases,19 cases were fallen into pathological grade Ⅰ,31 into grade Ⅱ,38 into grade Ⅲ,and 32 into grade Ⅳ.The patients with glioma under the standard treatment were followed up for 4 years.Results Pathological grade of glioma was increased with the increased expression of miRNA-221 and miRNA-222,and there were significant differences between the groups (x2 =43.53,P ＜0.01 ).Survival time was decreased with the increased expression of miRNA-221 and miRNA-222,and there were significant differences between the groups (x2 =55.63,P ＜ 0.0l ).Conclusion The expression of miRNA-221 and miRNA-222 was related to pathological grade and the prognosis of glioma,and can be considered as the independent prognostic factor.
