目的 检测PI3K、AKT、NRAS和ERK在色素痣和皮肤恶性黑素瘤组织中的表达情况,探讨它们的作用与意义. 方法应用SP免疫组织化学技术检测14例色素痣标本和11例皮肤恶性黑素瘤标本中PI3K 、AKT、NRAS和ERK蛋白的表达.结果 在皮肤恶性黑素瘤组,PI3K蛋白阳性表达率在皮肤恶黑组明显高于色素痣组(P<0.05);NRAS蛋白阳性表达率在皮肤恶性黑素瘤组明显高于色素痣组(P<0.05).AKT阳性表达率在皮肤恶性黑素瘤组和色素痣组比较无显著性差异(P>0.05);ERK阳性表达率在皮肤恶性黑素瘤组和色素痣组比较无显著性差异(P>0.05).在皮肤恶性黑素瘤中,四种蛋白间的表达没有相关性;在色素痣中四种蛋白间的表达亦没有相关性.结论 检测PI3K与NRAS的表达状况有助于皮肤恶性黑素瘤与色素痣的鉴别诊断.

Objective To explores the expression of the four sigaling molecules:PI3K,AKT,NRAS and ERK in cutaneous malignant melanoma and nevus pigmentosus,and to investigate their role and significance.Methods S-P method of immunohistochemistry was used to detect the expressions of PI3K,AKT,NRAS and ERK in 14 nevus pigmentosus specimens and 11 cutaneous malignant melanoma specimens.Results In cutaneous malignant melanoma,the positive expression rates of PI3K was significantly higher than the rate in nevus pigmentosus (P<0.05).The positive expression rates of NRAS in cutaneous malignant melanoma was significantly higher than the rate of nevus pigmentosus (P<0.05).There is no difference between the AKT positive rate of nevus pigmentosus and malignant melanoma(P>0.05).The positive expression rates of ERK in nevus pigmentosus showed no significant difference in cutaneous malignant melanoma (P>0.05).Positive expression of four kinds of proteinsin malignant melanoma have no correlation with each other(P>0.05),and as well as in nevus pigmentosus(P>0.05).Conclusion It could help to differentially diagnose cutaneous malignant melanoma from nevus pigmentosus by detecting the expression rate of PI3K and NRAS.
