将带有NPT-Ⅱ基因的质粒PGV用花粉管通道途径转化小麦烟农103,共结实1025粒,其中有718粒种子可以萌发出幼苗,在这718株幼苗中,经卡那霉素筛选,得到75棵绿色抗性植株,提取这些抗性植株的叶片总DNA,作NPT-Ⅱ基因的PCR扩增,有37株为阳性,转化率为5.16%,对其中9个PCR阳性植株的Southern杂交检测结果表明它们均为转基因植株.
將帶有NPT-Ⅱ基因的質粒PGV用花粉管通道途徑轉化小麥煙農103,共結實1025粒,其中有718粒種子可以萌髮齣幼苗,在這718株幼苗中,經卡那黴素篩選,得到75棵綠色抗性植株,提取這些抗性植株的葉片總DNA,作NPT-Ⅱ基因的PCR擴增,有37株為暘性,轉化率為5.16%,對其中9箇PCR暘性植株的Southern雜交檢測結果錶明它們均為轉基因植株.

The plasmid PGV with NPT-Ⅱ gene was transferred into wheat C. V.Yannnong 103 by pollen tube pathway. 1025 transformed seeds were selected with kanamycin,in which 718 germinated and 75 expressed resistence to kanamycin. Resistant green plants were determined by PCR amplificaton of transgenic fragment and confirmed by southern hybridization with a transformation efficiency of 5.16%.
