目的 检测胃癌组织及胃癌细胞中微小RNA (miRNA)表达谱,并测定胃癌组织及胃癌细胞中miR-429的表达水平.方法 采用基因芯片技术检测胃癌组织、癌旁正常胃组织标本及胃癌细胞株( SGC-7901)、正常人胃上皮细胞(GES-1)中miRNAs的表达.挑选基因芯片结果中胃癌组织表达显著下调的miR-429,并采用实时定量聚合酶链反应(qRT-PCR)检测7例正常胃组织、52例胃癌组织、胃癌细胞SGC-7901、GES-1中miR-429的表达.结果 基因芯片结果表明,胃癌组织中有23个miRNA上调,如miR-21、miR-30b等,31个下调如miR-429、miR-200等.qRT-PCR检测显示,miR-429在胃癌组织中下降约63.4% (P<0.01),在胃癌细胞株SGC7901中下降约76.2%(P<0.01).结论 miR-429在胃癌组织中表达显著下调,可能作为胃癌特异性miRNA,在胃癌的发生发展过程中有重要作用.

목적 검측위암조직급위암세포중미소RNA (miRNA)표체보,병측정위암조직급위암세포중miR-429적표체수평.방법 채용기인심편기술검측위암조직、암방정상위조직표본급위암세포주( SGC-7901)、정상인위상피세포(GES-1)중miRNAs적표체.도선기인심편결과중위암조직표체현저하조적miR-429,병채용실시정량취합매련반응(qRT-PCR)검측7례정상위조직、52례위암조직、위암세포SGC-7901、GES-1중miR-429적표체.결과 기인심편결과표명,위암조직중유23개miRNA상조,여miR-21、miR-30b등,31개하조여miR-429、miR-200등.qRT-PCR검측현시,miR-429재위암조직중하강약63.4% (P<0.01),재위암세포주SGC7901중하강약76.2%(P<0.01).결론 miR-429재위암조직중표체현저하조,가능작위위암특이성miRNA,재위암적발생발전과정중유중요작용.
Objective To detect the expression profiling of microRNA (miRNA) and the expression level of miRNA-429 in human gastric carcinoma.Methods The miRNAs expression of 7 normal gastric tissues,52 malignant tissues,gastric cancer cell line SGC7901 and gastric epithelial cell line 1 ( GES-1 ) was detected by using miRNA gene chips.Real-time reverse transcriptase-polymerase chain reaction (qRT-PCR) was used to quantify the expression level of miR-429 in 52 gastric cancer tissues and their paracancerous tissues.Results The results of gene chip showed that 23 miRNA were significantly up-regulated,such as miR-21,miR-30b,etc.,and 31 miRNA down-regulated,such as miR-429,miR-200,etc.The average expression of miR-429 was significantly reduced by 63.4% in gastric cancer samples as compared with that in normal paracancerous tissues (P <0.01 ).miR-429 expression was reduced by 76.2%in SGC-7901 cells as compared with non-tumor gastric cells (P <0.01 ).Conclusion miR-429 might play a role in the pathogenesis of gastric carcinoma.
