目的 探讨基质金属蛋白酶(MMPs) 在类风湿关节炎(RA)和系统性红斑狼疮(SLE)中的重要作用.方法 采用酶联免疫吸附试验和酶谱分析了48例RA和27例SLE患者及186例健康对照者血清中MMP-2和MMP-9的浓度和活性.结果 酶谱分析结果 显示RA和SLE患者血清MMP-2和MMP-9的活性显著高于对照组(P<0.01).ELISA检测结果 也表明RA、SLE组的MMP-2和MMP-9 浓度明显高于对照组(P<0.05).结论 MMP-2和MMP-9在RA和SLE患者血清中的水平及活性明显升高,表明MMPs在这些自身免疫疾病起一定的作用,可作为临床辅助诊断指标.

Objective To investigate the importance of matrix metalloproteinases (MMPs) in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) and determine the MMPs concentrations and activities in RA and SLE patients. Methods The levels and activities of MMP-2 and MMP-9 were detected in serum of 48 RA and 27 SLE patients and 186 healthy controls by enzyme-linked immunosorbent assay (ELISA) and zymography. Results The results of zymography showed that the activities of MMP-2 and MMP-9 in the RA and SLE patients were significantly higher than those of healthy controls(P<0.01). The results of ELISA also showed that the concentrations of MMP-2 and MMP-9 in the RA and SLE patients were significantly higher than those of healthy controls(P<0.05). Conclusions The results of MMP-2 and MMP-9 levels and activities significantly increase, which shows that MMPs plays an important role in these autoimmune diseases. They may become the auxiliary diagnose index in clinic.
