目的 观察糖尿病肾病(DN)大鼠肾组织c-Ski表达,探讨其在肾小管上皮向肌成纤维细胞转分化(TEMT)中的意义.方法 36只Wistar雄性大鼠随机分为正常组(n=12)、模型组(n=12)和干预组[大黄酸100 mg/(kg·d),n=12].第16周处死大鼠,观察大鼠肾脏病理改变并检测肾组织中转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)、c-Ski的表达.结果 DN大鼠肾小管上皮细胞c-Ski表达与TGF-β1、α-SMA表达呈负相关;干预组大鼠肾小管上皮细胞c-Ski表达较模型组高.结论 c-Ski是TEMT的一个重要保护性因子.
目的 觀察糖尿病腎病(DN)大鼠腎組織c-Ski錶達,探討其在腎小管上皮嚮肌成纖維細胞轉分化(TEMT)中的意義.方法 36隻Wistar雄性大鼠隨機分為正常組(n=12)、模型組(n=12)和榦預組[大黃痠100 mg/(kg·d),n=12].第16週處死大鼠,觀察大鼠腎髒病理改變併檢測腎組織中轉化生長因子-β1(TGF-β1)、α-平滑肌肌動蛋白(α-SMA)、c-Ski的錶達.結果 DN大鼠腎小管上皮細胞c-Ski錶達與TGF-β1、α-SMA錶達呈負相關;榦預組大鼠腎小管上皮細胞c-Ski錶達較模型組高.結論 c-Ski是TEMT的一箇重要保護性因子.
목적 관찰당뇨병신병(DN)대서신조직c-Ski표체,탐토기재신소관상피향기성섬유세포전분화(TEMT)중적의의.방법 36지Wistar웅성대서수궤분위정상조(n=12)、모형조(n=12)화간예조[대황산100 mg/(kg·d),n=12].제16주처사대서,관찰대서신장병리개변병검측신조직중전화생장인자-β1(TGF-β1)、α-평활기기동단백(α-SMA)、c-Ski적표체.결과 DN대서신소관상피세포c-Ski표체여TGF-β1、α-SMA표체정부상관;간예조대서신소관상피세포c-Ski표체교모형조고.결론 c-Ski시TEMT적일개중요보호성인자.
Objective To observe the expressions of c-Ski in renal tissue of diabetic nephropathy rats, and discuss its significance. Methods Wistar male rats were randomly assigned to 3 groups: Control group, diabetic group and treatment group. Rats were killed at the 16th week after experiment. Histopathologic changes,in renal tissue were observed and immunohistochemistry was performed to investigate the expression of TGF-β1, α-SMA and c-Ski. Results The expression of c-Ski had a negative correlation with TGF-β1 and α-SMA. Compared to diabetic group, the expression of c-Ski was significantly increased in treatment group. Conclusion c-Ski may be a protective factor of tu-bular epithelial-myofibroblast transformation.
