目的:利用RNA干扰(RNA interference,RNAi)技术在裸鼠皮下移植瘤中抑制 Livin 基因表达,探讨 RNA 干扰用于胃癌治疗的可行性.方法:SGC-7901 细胞注射入裸鼠皮下,建立裸鼠皮下移植瘤的模型,设计靶向Livin基因的小干扰RNA(small interference RNA,siRNA),构建重组表达质粒 pTZU6+I-siRNA-Livin,并导入裸鼠皮下移植瘤中,在体内诱导RNAi,采用RT-PCR和免疫化学检测Livin基因表达,Western blot 检测 Bax 和B cl-2 改变.结果:重组质粒 pTZU6+1-siRNA-Livin 导入裸鼠皮下移植瘤后14 d 瘤体体积明显减小,Livin 的 mRNA 和蛋白表达均明显下调.结论:RNA 干扰明显抑制了靶基因 Livin 的表达及肿瘤细胞生长,可能与上调Bax的表达、下调 Bcl-2,从而诱导肿瘤细胞凋亡有关.

목적:이용RNA간우(RNA interference,RNAi)기술재라서피하이식류중억제 Livin 기인표체,탐토 RNA 간우용우위암치료적가행성.방법:SGC-7901 세포주사입라서피하,건립라서피하이식류적모형,설계파향Livin기인적소간우RNA(small interference RNA,siRNA),구건중조표체질립 pTZU6+I-siRNA-Livin,병도입라서피하이식류중,재체내유도RNAi,채용RT-PCR화면역화학검측Livin기인표체,Western blot 검측 Bax 화B cl-2 개변.결과:중조질립 pTZU6+1-siRNA-Livin 도입라서피하이식류후14 d 류체체적명현감소,Livin 적 mRNA 화단백표체균명현하조.결론:RNA 간우명현억제료파기인 Livin 적표체급종류세포생장,가능여상조Bax적표체、하조 Bcl-2,종이유도종류세포조망유관.
Objective:To investigate the feasibility of gastric carcinoma gene therapy by utilizing RNA interference (RNAi) to inhibit the expression of Livin in vivo.Methods:SGC-7901was injected into nude mice to form hypodermical gastric carcinoma.Small interference RNA(siRNA) homologous to Livin gene were designed,pTZU6+1-siRNA-Livin vector was constructed and injected into tumor to induce RNAi.The changes of Livin gene expression were detected with RT-PCR and immunochemistry,the expression of Bax and Bcl-2 were detected by western blot.Results:14 days after transfected with pTZU6+1-siRNA-Livin,both the mRNA and the protein of Livin decreased obviously.The level of Bax was up-regulated and the level of Bcl-2 was decreased.Conclusion:RNA interference targeting Livin inhibits Livin expression and tumor growth,which may be related to up-regnlation of Bax and down-regulation of Bcl-2.
