用人胃癌细胞株AGS作为抗原免疫Balb/c小鼠,通过杂交瘤技术制备抗人胃癌细胞mAb,应用间接ELISA法进行筛选,显微操作法进行亚克隆并用琼脂糖双向免疫扩散法鉴定亚类,免疫化学法检测特异性,获得l株分泌抗AGS细胞株的mAb的杂交瘤(命名为1B4).该杂瘤分泌的mAb与人宫颈癌细胞株Hela、肝癌细胞株HepG2和正常胎儿肺细胞均为阴性反应,而与AGS细胞呈阳性反应.结果表明,该mAb对AGS细胞株具有一定的特异性,可为胃癌相关抗原的筛选研究提供一定的帮助.

Balb/c mice were immunized with human gastric adenocarcinoma cell line AGS and the B lymphocytes hybridoma technique was used for mAb preparation.The property of antibody was studied with indirect ELISA,double immunodiffusion and immunocytochemical method to identify the specific reaction.A hybridoma cell line,1B4,which secretes mAb against AGS cell line was established.The mAb could recognize an AGS cell line but not HeLa,HepG2 and human fetal lung cell lines.The results suggested that the mAb might be specific to AGS and be useful for the research on human gastric cancer-associated antigens.
