目的:诱变筛选阿魏菇多糖高产菌,探索食用菌的离子束和激光复合诱变育种方法.方法:尝试用阿魏菇菌丝单细胞为靶材,以离子束注人和激光辐照为复合诱变手段,采用营养缺陷型筛选办法定性初筛,摇瓶发酵定量复筛.结果:通过离子束注入诱变,获得了2株菌丝体多糖产量分别达到551.80mg/L和659.46mg/L、较l号出发菌株提高了46.5%和75.2%的多糖高产菌PFPH-1和PFPH-2;在此基础上,以激光辐照为复合诱变手段,获得了1株菌丝体多糖产量达到762.50 mg/L、较出发菌株PFPH-2提高了15.63%的多糖高产菌PFPH-3.结论:离子束和激光复合诱变育种方法在改良阿魏菇多糖高产性状方面诱变功效显著.
目的:誘變篩選阿魏菇多糖高產菌,探索食用菌的離子束和激光複閤誘變育種方法.方法:嘗試用阿魏菇菌絲單細胞為靶材,以離子束註人和激光輻照為複閤誘變手段,採用營養缺陷型篩選辦法定性初篩,搖瓶髮酵定量複篩.結果:通過離子束註入誘變,穫得瞭2株菌絲體多糖產量分彆達到551.80mg/L和659.46mg/L、較l號齣髮菌株提高瞭46.5%和75.2%的多糖高產菌PFPH-1和PFPH-2;在此基礎上,以激光輻照為複閤誘變手段,穫得瞭1株菌絲體多糖產量達到762.50 mg/L、較齣髮菌株PFPH-2提高瞭15.63%的多糖高產菌PFPH-3.結論:離子束和激光複閤誘變育種方法在改良阿魏菇多糖高產性狀方麵誘變功效顯著.

Objective:To obtain Pleurotus polysaccharide high - yield strains and explore an ion beam and laser composite mutagenesis breeding method for edible fungus. Method:Mycelium mono - cells of Asafoetida mushroom were treated tentatively by ion beam and laser composite mutagenesis through fermentation quantitative screening after auxotrophy qualitative primary screening. Result:Two polysaccharide high - y ield strains, PFPH - 1 and PFPH - 2, were selected by ion beam implantation. The polysaccharide yield of these mutants was 551.80mg/L and 659.46mg/L, which were increased by 46.5% and 75.2% compared to that of initial strain No.1. On this basis, a polysaccharid e high - yield strain named PFPH - 3 was obtained by laser eradiation as composite mutagenesis method. The polysaccharide was 762.50mg/L, which was increased by 15.63% compared to that of initial strain PFPH -2. Conclusion: In this study, the mutagenic effect was significant that ion beam and laser composite mutagenesis breeding method was used in improving Pleurotus polysaccharide yield.
