目的 探讨多囊卵巢综合征(PCOS)患者与正常排卵妇女子宫内膜脂联素(APN)及其受体(AdipoR)表达的差异.方法 选择体质指数<25 kg/m~2的PCOS患者(PCOS组)和正常排卵妇女(对照组)各10例,收集月经第10~12天的子宫内膜.通过蛋白印迹和实时定量聚合酶链反应(PCR)等方法测定子宫内膜APN及AdipoR1、AdipoR2的表达.结果 ①蛋白印迹结果显示,PCOS组与对照组间子宫内膜APN表达的差异无统计学意义(P>0.05),两组患者的子宫内膜均有微弱的AdipoR1及AdipoR2表达.②实时定量PCR结果显示,PCOS与对照组的APN mRNA、AdipoR1 mRNA及AdipoR2 mRNA表达的差异均无统计学意义(P值均>0.05).PCOS组的AdipoR1 mRNA与AdipoR2 mRNA表达的差异无统计学意义(P>0.05),对照组的AdipoR2 mRNA表达则显著低于AdipoR1 mRNA(P<0.05).结论 体重正常的PCOS患者的子宫内膜无APN及其受体表达水平异常.

Objective To compare the expressions of adiponectin (APN), AdipoR1 and AdipoR2 in endometrial tissues between patients with polycystic ovary syndrome (PCOS) and normally ovulating women. Methods The proliferative phase endometrial tissues were collected from 10 infertile women with PCOS and 10 normally ovulating women. Expressions of adiponectin, AdipoR1 and AdipoR2 in the endometrial tissues were examined by Western blotting analysis and real-time quantitative PCR. Results ① Western blotting analysis showed that the expressions of APN were not significantly different between the two groups (P>0.05), and weak expressions of AdipoR1 and AdipoR2 were seen in the endometrial tissues of both groups. ② Real-time PCR demonstrated that the expressions of APN mRNA, AdipoR1 mRNA, and AdipoR2 mRNA were not significantly different between the two groups (all P>0. 05). The expressions of AdipoR1 mRNA and AdipoR2 mRNA were similar in PCOS group (P>0.05), and the expression of AdipoR2 mRNA was significantly lower than that of AdipoR2 mRNA in the control group (P<0.05). Conclusion It is suggested PCOS patients with normal weight have no abnormal expression of adiponectin and its receptors in the endometrium.
