目的 观察胰岛素样生长因子-1(IGF-1)基因转染的骨髓间质干细胞(BMSCs)移植对糖尿病大鼠骨折愈合的影响,探讨糖尿病骨折的基因疗法.方法 Wistar雄性6周龄大鼠50只,随机分为对照组、实验组,链脲佐菌素诱导为糖尿病模型后均造成右侧胫骨骨折,体外高糖环境下Ad-IGF-1转染BMSCs,将相应组别的BMSCs移植于骨折局部.于1、2、3、4、6周摄X线,取局部骨痂行苏木素-伊红(HE)染色,并免疫组织化学检测骨痂中IGF-1,酶联免疫吸附试验(ELISA)检测血清IGF-1.结果 第4周骨痂IGF-1灰度值:实验组为103±5,对照组为109±4(P<0.05).第4周血清IGF-1浓度:实验组为(668.80±8.07) ng/L,对照组为(569.20±9.31) ng/L(P <0.01).结论 IGF-1基因转染的BMSCs移植能促进糖尿病大鼠的骨折愈合.
目的 觀察胰島素樣生長因子-1(IGF-1)基因轉染的骨髓間質榦細胞(BMSCs)移植對糖尿病大鼠骨摺愈閤的影響,探討糖尿病骨摺的基因療法.方法 Wistar雄性6週齡大鼠50隻,隨機分為對照組、實驗組,鏈脲佐菌素誘導為糖尿病模型後均造成右側脛骨骨摺,體外高糖環境下Ad-IGF-1轉染BMSCs,將相應組彆的BMSCs移植于骨摺跼部.于1、2、3、4、6週攝X線,取跼部骨痂行囌木素-伊紅(HE)染色,併免疫組織化學檢測骨痂中IGF-1,酶聯免疫吸附試驗(ELISA)檢測血清IGF-1.結果 第4週骨痂IGF-1灰度值:實驗組為103±5,對照組為109±4(P<0.05).第4週血清IGF-1濃度:實驗組為(668.80±8.07) ng/L,對照組為(569.20±9.31) ng/L(P <0.01).結論 IGF-1基因轉染的BMSCs移植能促進糖尿病大鼠的骨摺愈閤.
목적 관찰이도소양생장인자-1(IGF-1)기인전염적골수간질간세포(BMSCs)이식대당뇨병대서골절유합적영향,탐토당뇨병골절적기인요법.방법 Wistar웅성6주령대서50지,수궤분위대조조、실험조,련뇨좌균소유도위당뇨병모형후균조성우측경골골절,체외고당배경하Ad-IGF-1전염BMSCs,장상응조별적BMSCs이식우골절국부.우1、2、3、4、6주섭X선,취국부골가행소목소-이홍(HE)염색,병면역조직화학검측골가중IGF-1,매련면역흡부시험(ELISA)검측혈청IGF-1.결과 제4주골가IGF-1회도치:실험조위103±5,대조조위109±4(P<0.05).제4주혈청IGF-1농도:실험조위(668.80±8.07) ng/L,대조조위(569.20±9.31) ng/L(P <0.01).결론 IGF-1기인전염적BMSCs이식능촉진당뇨병대서적골절유합.
Objective To observe the effect of transplantation of bone marrow mesenchymal stem cells (BMSCs) transfected by insulin-like growth factor-1 ( IGF-1 ) gene on fracture healing of diabetic rats.Methods Fifty male Wistar rats were divided randomly into control group and transfection group.Streptozotocin was injected into celiac to induce diabetes model and all suffered right tibia fracture.Ad-IGF-1 transfected rat BMSCs under high glucose condition in vitro. After transplantation of BMSCs transfected by IGF-1,X-ray photographs were taken at 1,2,3,4,6 weeks,and the callus was subjected to hematoxylin and eosin (HE) staining.The levels of IGF-1 of the callus and serum were detected by immunohistochemical method and enzyme linked immunosorbent assay (ELISA) respectively. Results There were significant differences in the expression of the IGF-1 gene between the two groups.At 4 weeks,the gray scales of IGF-1 were 103 ±5 in transfection group and 109 ±4 in control group (P <0.05).The serum concentrations of IGF-1 at 4 weeks were (668.80 ±8.07) ng/L in transfection group and (569.20 ±9.31 ) ng/L in control group (P <0.01 ).Conclusion The transplantation of BMSCs transfected by IGF-1gene can promote the fracture healing of diabetic rats.
