目的 分析肾细胞癌肿瘤特异性蛋白质,寻找新的肿瘤分子标志物.方法 应用二维电泳(2D-PAGE)分析9例肾细胞癌患者癌组织和周边的正常肾组织中差异表达的蛋白分子,并运用MALDI-TOF-MS和MS/MS法对差异表达的蛋白质标记点进行了鉴定.结果 经过二维电泳分离样本,发现了差异表达的16个蛋白质点(P<0.05,t检验),用MALDI-TOF-MS法对这些点进行分析鉴定,确定了他们的种类,其中表达下调的有钙结合蛋白、酯类水解酶、乙醇脱氢酶、ammecr-1样蛋白、ADP/ATP转位-3和LYRM-5;表达上调的有HIG-1、谷胱甘肽S.转移酶(GSTP-1)、Peroxiredoxin-3、Peroxiredoxin-6、CD2相关蛋白(CD2AP)、Annexin V、γ-烯醇化酶、视网膜脱氢酶、波形蛋白(vimentin)和α-谷氨酰转移酶.2.结论 在筛选到的16种蛋白质中,钙结合蛋白、ADP/ATP转位酶-3、HIG-1、谷胱甘肽-S-转移酶、Peroxiredoxin-3和6是比较有研究价值的分子.

Objective To identify tumor specific proteins of renal cell carcinoma(RCC)by using proteomic analysis.Methods The expression of proteins was detected by silver stain after two-dimensional polyacrylamide gel electrophoresis(2D-PAGE).The differentially expressed proteins were identified by MALDI-TOF-MS.Results Six proteins were down-regulated and 10 proteins up-regulated in RCC tissues as compared with normal kidney tissues by silver stain after 2D-PAGE.The 16 differentially expressed protein spots(P<0.05.by Student t-test)were identified by MALDl-TOF-MS and MS/MS.The down-regulated proteins included calbindin,ester hydrolase,alcohol dehydrogenase,ammecr-1-like protein,ADP/ATP tranglocase-3 and LYR motif-containing protein-5.and the up-regulated proteins included HIC-1 domain family member 1 A,glutathione-S-transferaae-P,thioredoxin-dependent-peroxide-reduetase,peroxiredoxin-6,CD2-associated protein,Annexin A5,gamma-enolase,retinal dehydrogenase-1,vimentin,and protein-gluramine gamma-glutamyltransferase-2.Conclusion Among the 16 proteins,Calbindin,ADP/ATP translocase3.HIG-1.glutathione-S-transferase-P-1,and thioredoxin-dependent-peroxide-reductase were worthy of further investigation.and maybe they will be developed into diagnostic markers for RCC.
