目的检测肾癌中SPINT2基因启动子甲基化状态,探讨其与肿瘤特征及患者预后的关系.方法选取2002～2004年本我院就诊的肾透明细胞癌石蜡组织,用甲基化特异性PCR检测其中SPINT2基因启动子甲基化状态,分析其与肿瘤特征及患者预后的关系.结果共42例肾透明细胞癌患者密切随访,平均随访39个月(3 ～78个月).其中完全甲基化19例,不完全甲基化13例,无甲基化10例.不同甲基化状态的患者预后存在显著性差异(P=0.042),甲基化与肿瘤分期显著相关(P=0.007).结论 SPINT2基因启动子甲基化在肾透明细胞癌组织中广泛存在,其甲基化状态与肾癌的肿瘤分期和预后密切相关.

Objectives To investigate the aberrant methylation of SPINT2 in renal carcinoma and its correlation to prognosis.Methods The promoter methylation status of 42 cases of renal clear cell carcinoma,which were treated in our center from 2002 to 2004,was tested by methylation-specific PCR.Correlations between methylation status and prognosis or clinical characters were analyzed.Results 42 cases of patients met our criteria,with median follow-up of 39 months (3～78 months).Total methylated SPINT2 gene were detected in 19 cases,partial methylated SPINT2 gene were detected in 13 cases and un-methylated were detected in 10 cases.Methylation of SPINT2 gene is related to clinical stage and prognosis ( P =0.007&0.042,respectively).Conclusions Methylation of SPINT2 gene is expressed wide-spread in renal clear cell carcinoma,and related to clinical stage and prognosis.Hypermethylation may play an important role in renal carcinoma.
