建立了八角中莽草酸的毛细管区带电泳-紫外检测分析新方法.在单一磷酸盐和硼砂溶液中由于电扩散现象,莽草酸峰宽并严重前倾或脱尾,采用磷酸盐-硼砂混合溶液为缓冲液改善了峰形并提高了检测灵敏度,通过优化获得其最佳分离条件为pH 8.63,10 mmol/L NaH2PO4-9 mmol/L硼砂,分离电压25 kV.该条件下莽草酸的峰面积与浓度呈良好的线性关系,线性方程的相关系数为0.9998,检测限(S/N=3)为0.12μg/mL,比文献报道的方法约高10倍.该方法成功用于八角壳和籽中莽草酸的定量分析,其加标回收率分别为98.9%和104.6%.

A simple and rapid capillary zone electrophoresis method using phosphate-borate mixture as running electrolyte with direct UV detection was developed for the analysis of shikimic acid.In pure phosphate and borate buffer,electro-dispersion of sample zone leads to peak leading and tailing because the motilities have the relationship as borate>shikimic acid>phosphate.Using a mixture of phosphate and borate as buffer,the peak efficiency and adsorption signal of shikimic acid axe much improved.After optimization,the separation conditions are selected as 10 mmol/L NaH2PO4+9 mmol/L Na2B4O7 solution,pH 8.63,separation voltage 25 kV.Under this condition,good linear relationship (R=0.999 8) between peak area and the concentration of shikimic acid is revealed in the tested range and the limit of detection is determined to be 0.12 μg/mL,which is about ten times lower than reported values in literatures.Finally the method was applied to determine the contents of shikimic acid in the shell and core of Illicium verum Hook F.with recoveries of 98.9% and 104.6% respectively.
