本研究利用限制性内切酶片段长度多态性(RFLP)和16S rDNA序列分析相结合的方法研究了广西南宁高峰林场人工桉树林中土壤微生物的群落结构.通过采用直接法提取桉树人工林土壤中微生物总DNA,构建细菌的16S rDNA克隆文库,从中随机挑取了192个阳性克隆进行检测,分析显示188个阳性克隆子确定含有16S rDNA并分归为52个不同的类群OTUs,随机挑取其中35个克隆进行测序,并构建了系统发育进化树.研究结果表明:桉树人工林土壤中细菌种类较为丰富,含有大量的未培养微生物,进一步的序列分析表明桉树人工林土壤中占优势的类群分别为Acidobacteria(40%)和Proteobacteria(27%).本研究对桉树人工林土壤微生物群落结构进行了研究,为进一步研究桉树人工林环境质量变化提供基础资料.

In this research,we investigated the edaphon community structure from Gaofeng forest-farm Eucalyptus plantation in Guangxi Nanning by PCR-RFLP technique and 16S rDNA sequence analysis.Total DNA of microorganism in Eucalyptus plantation was extracted by using the direct method,then we constructed a 16S rDNA clone library of the soil microbes.A total of 192 positive clones were radomly chosen from the clone library to be examined.The PCR-RFLP result showed that 188 positive clones determinately contained 16S rDNA and they went back to 52 different operational taxonomic units (OTUs).We also radomly chose 35 clones among them to sequence and constructed phylogenetic development tree.The results indicated that the bacteria species in Eucalyptus plantation soil are relatively abundance.Further sequence analysis showed that the dominant bacteria in this Eucalyptus plantation soil were A cidobacteria and Proteobacteria which was 40% and 27%,respectively.This resarech investigated the edaphon community structure of Eucalyptus plantation,which would provide the foundation information for studying the variety in environmental quality of Eucalyptus plantation.
