目的检测CXC趋化因子受体4(CXCR4)mRNA及其蛋白在舌癌组织及转移颈淋巴结组织中的表达水平,并探讨其在舌癌预后和颈淋巴结转移中的作用.方法采用免疫组化SABC法检测41例舌癌组织及41例颈部淋巴结标本中CXCR4的表达及定位,用半定量RT-PCR检测各标本CXCR4的mRNA相对表达值.结果免疫组化结果显示,舌癌原发灶和颈淋巴结转移灶表达CXCR4,正常舌组织不表达CXCR4或仅呈弱阳性表达,正常淋巴结不表达CXCR4;RT-PCR结果显示,舌癌以及转移淋巴结表达CXCR4 mRNA,正常舌组织和正常淋巴结不表达或低表达CXCR4 mRNA.结论舌癌组织及转移颈淋巴结中有CXCR4 mRNA及蛋白的表达,其表达与舌癌的预后及颈淋巴结的转移有关.

Objective To detect the mRNA and protein level of CXCR4 in human tongue carcinoma and metastatic cervical lymph nodes. Methods The mRNA of CXCR4 in the human tongue carcinoma and metastatic cervical lymph node from 41 paient was tested by semi-quantitative RT-PCR. Strept Avidin-Biotin-Complex Method (SABC Method) was employed to delect the localization, intensity and distribution of CXCR4 protein. Results Immunohistochemistry staining and RT-PCR demonstrated that CXCR4 was expressed in tongue carcinoma and metastatic cervical lymph nodes, and was not or just weakly expressed in normal tounge tissue and lymph nodes. Conclusions The mRNA and protein of CXCR4 tongue carcinoma and metastatic cervical lymph node. The expressions of CXCR4 is correlated with the prognosis and cervical lymph node metastasis of tongue carcinoma.
