目的:采用改进的离子对高效液相色谱方法测定人血浆中二甲双胍的浓度.方法:乙腈作为蛋白沉淀剂,除去酸化血浆样品中的蛋白,二氯甲烷进一步去杂质后,取上清液进样.流动相为甲醇(56%),0.35 mmol·L-1的十二烷基硫酸钠和5mmol·L-1的磷酸盐缓冲液(44%,pH 6.6),流速为1.2mL·min-1,紫外检测波长234 nm.结果:二甲双胍和内标(哌仑西平)的保留时间分别为7.1和4.8 min,标准曲线在20～4 000mg·L-1浓度范围内线性关系良好,绝对回收率＞95%,检测限和定量限分别为4和10 mg·L-1.天内和天间RSD＜6.4%.结论:该方法适用于盐酸二甲双胍的治疗药物监测及临床药学研究.

AIM: To develop a ion pair liquid chromatographic method for specific HPLC determination of metformin hydrochloride. METHODS: Acidified samples of plasma were deproteinated with acetonitrile, washed with at 234 nm. RESULTS: The retention time of metformin hydrochloride and the internal standard was eluted 7.1 and 4.8recovery of metformin hydrochloride and the internal standard was greater than 95 %. The detection limit and limit of CLUSION: This method is suitable for therapeutic drug monitoring and clinical investigation of metformin hydrochloride.
