目的确定柠檬酸铵在紫杉醇生物代谢途径的作用位点。方法分别在0,0.1,1,10,20μmol/L的柠檬酸铵诱导下,观察紫杉烷类物质产量的变化,并通过生物代谢途径进行定性分析。结果紫杉醇的产量提高近3倍,而10-DAB(10-deactylbaccatin Ⅲ)与baccatin Ⅲ浓度下降。结论柠檬酸铵提高了baccatin Ⅲ与紫杉醇间的反应速度,即其作用位点位于baccatin Ⅲ与紫杉醇之间。
目的確定檸檬痠銨在紫杉醇生物代謝途徑的作用位點。方法分彆在0,0.1,1,10,20μmol/L的檸檬痠銨誘導下,觀察紫杉烷類物質產量的變化,併通過生物代謝途徑進行定性分析。結果紫杉醇的產量提高近3倍,而10-DAB(10-deactylbaccatin Ⅲ)與baccatin Ⅲ濃度下降。結論檸檬痠銨提高瞭baccatin Ⅲ與紫杉醇間的反應速度,即其作用位點位于baccatin Ⅲ與紫杉醇之間。
목적학정저몽산안재자삼순생물대사도경적작용위점。방법분별재0,0.1,1,10,20μmol/L적저몽산안유도하,관찰자삼완류물질산량적변화,병통과생물대사도경진행정성분석。결과자삼순적산량제고근3배,이10-DAB(10-deactylbaccatin Ⅲ)여baccatin Ⅲ농도하강。결론저몽산안제고료baccatin Ⅲ여자삼순간적반응속도,즉기작용위점위우baccatin Ⅲ여자삼순지간。
Object To study the eliciting effec t of ammonium citrate (AC) onthe yield of taxol. Methods The study was carried out on plant cell cultures with different concentrations of ad ded AC. Results Yield of taxol was increased by 3 times under 10 μmol/L AC elicitation, while the concentrations of 10-deacetyl baccatin Ⅲ and baccatin Ⅲ were simultaneously decreased. Conclusion By a nalyzing the changing characters between substrate and enzyme in the simplified taxol synthetic pathway. AC was considered to play the role in increasing the reaction rate from baccatin Ⅲ to taxol.
