目的 观察人羊膜间充质干细胞( hAD - MSCs)移植对大鼠脊髓损伤神经功能恢复的影响.方法 建立大鼠脊髓全横断损伤模型,脊髓横断后立即以明胶海绵吸附10μl hAD- MSCs(约2×105个)或等量PBS液植入脊髓两断端之间.术后每周应用BBB评分评价大鼠后肢运动功能;采用免疫荧光染色观察hAD - MSCs在脊髓内的存活、分化情况;免疫组织化学染色观察受损脊髓远端组织NF - 200表达.结果 hAD - MSCs移植组神经功能明显恢复,BBB评分逐周增加,与对照组比较差异有统计学意义(P<0.05).hAD - MSCs植入后2周在宿主脊髓中存在MAB1281染色阳性细胞,但不表达MAP -2和GFAP.hAD - MSCs移植后大鼠受损脊髓远端神经组织NF - 200表达明显强于对照组.结论 hAD - MSCs移植可促进大鼠脊髓损伤后的神经功能恢复,其机制可能与hAD - MSCs促进受损脊髓远端组织表达NF - 200有关.
目的 觀察人羊膜間充質榦細胞( hAD - MSCs)移植對大鼠脊髓損傷神經功能恢複的影響.方法 建立大鼠脊髓全橫斷損傷模型,脊髓橫斷後立即以明膠海綿吸附10μl hAD- MSCs(約2×105箇)或等量PBS液植入脊髓兩斷耑之間.術後每週應用BBB評分評價大鼠後肢運動功能;採用免疫熒光染色觀察hAD - MSCs在脊髓內的存活、分化情況;免疫組織化學染色觀察受損脊髓遠耑組織NF - 200錶達.結果 hAD - MSCs移植組神經功能明顯恢複,BBB評分逐週增加,與對照組比較差異有統計學意義(P<0.05).hAD - MSCs植入後2週在宿主脊髓中存在MAB1281染色暘性細胞,但不錶達MAP -2和GFAP.hAD - MSCs移植後大鼠受損脊髓遠耑神經組織NF - 200錶達明顯彊于對照組.結論 hAD - MSCs移植可促進大鼠脊髓損傷後的神經功能恢複,其機製可能與hAD - MSCs促進受損脊髓遠耑組織錶達NF - 200有關.
목적 관찰인양막간충질간세포( hAD - MSCs)이식대대서척수손상신경공능회복적영향.방법 건립대서척수전횡단손상모형,척수횡단후립즉이명효해면흡부10μl hAD- MSCs(약2×105개)혹등량PBS액식입척수량단단지간.술후매주응용BBB평분평개대서후지운동공능;채용면역형광염색관찰hAD - MSCs재척수내적존활、분화정황;면역조직화학염색관찰수손척수원단조직NF - 200표체.결과 hAD - MSCs이식조신경공능명현회복,BBB평분축주증가,여대조조비교차이유통계학의의(P<0.05).hAD - MSCs식입후2주재숙주척수중존재MAB1281염색양성세포,단불표체MAP -2화GFAP.hAD - MSCs이식후대서수손척수원단신경조직NF - 200표체명현강우대조조.결론 hAD - MSCs이식가촉진대서척수손상후적신경공능회복,기궤제가능여hAD - MSCs촉진수손척수원단조직표체NF - 200유관.
Objective To study the effects of transplantation of human aminon -derived mesenchymal stem cells on the neurological function restoration of rats with spinal cord injury.Methods Transected spinal cord injury(SCI) model in rats was established.hAD - MSCs (about 2 × 105 ) or equal volume of PBS absorbed by a gelfoam were transplanted into the interval of injury ends immediately after SCI.The movement function of hindlimbs was evaluated by Basso-Beattie-Bresnahan(BBB) scorse every week after SCI.The survival and differentiation of hAD-MSCs in the injured spinal cord were evaluated by immunofluorescence staining.The expression of NF-200 in the distal end of injured site was detected with immunohistochemical staining.Results The neurological function in hAD- MSCs transplantation group was improved significantly and the BBB score was increased gradually ( P <0.05).The grafting hAD -MSCs were MAB1281postively at the 2nd week after transplantation,but were not MAP-2 and GFAP postively.The expression of NF -200 in the distal end of injured site of spinal cord of hAD -MSCs transplantation group was remarkably higher than that of PBS control group after transplantation.Condusions Transplantation of hAD - MSCs could be benefit to improve neurological function restoration of rats with SCI.Its mechanism might relate to the promotion of NF- 200 expression in the distal end of injured spinal cord after hAD-MSCs transplantation.
