目的 探讨鱼油对开放式腹部手术大鼠术后疲劳的影响及机制.方法 将24只SD大鼠均分成对照组(C组)、手术组(S组)和鱼油干预组(F组).术后观察行为学变化,检测白细胞介素(IL)-1β、IL-6、肿瘤坏死因子(TNF)-α及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)含量;光镜观察脑海马组织形态,电镜观察其超微结构;高效液相检测中枢神经递质去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT).结果 病理和电镜显示F组损伤较S组轻;术后高效液相分析表明中枢不同区域的神经递质含量不同.S组血清IL-1β、IL-6、TNF-α浓度分别为(60.02±11.08)、(50.13 ±4.45)、(41.08±2.17) ng/L,F组为(45.48±13.51)、(42.08±4.47)、(38.76±2.41) ng/L,均较C组增高(P<0.01),F组较S组则明显降低(P<0.01);F组的血清SOD、GSH-PX含量较S组高(P<0.01);F组Morris水迷宫实验指标优于S组(P<0.05).结论 鱼油能有效改善术后的炎症反应,减轻抗氧化防御系统的损伤,减轻中枢对外周手术创伤的应激,改善术后疲劳.

목적 탐토어유대개방식복부수술대서술후피로적영향급궤제.방법 장24지SD대서균분성대조조(C조)、수술조(S조)화어유간예조(F조).술후관찰행위학변화,검측백세포개소(IL)-1β、IL-6、종류배사인자(TNF)-α급초양화물기화매(SOD)、곡광감태과양화물매(GSH-PX)함량;광경관찰뇌해마조직형태,전경관찰기초미결구;고효액상검측중추신경체질거갑신상선소(NE)、다파알(DA)、5-간색알(5-HT).결과 병리화전경현시F조손상교S조경;술후고효액상분석표명중추불동구역적신경체질함량불동.S조혈청IL-1β、IL-6、TNF-α농도분별위(60.02±11.08)、(50.13 ±4.45)、(41.08±2.17) ng/L,F조위(45.48±13.51)、(42.08±4.47)、(38.76±2.41) ng/L,균교C조증고(P<0.01),F조교S조칙명현강저(P<0.01);F조적혈청SOD、GSH-PX함량교S조고(P<0.01);F조Morris수미궁실험지표우우S조(P<0.05).결론 어유능유효개선술후적염증반응,감경항양화방어계통적손상,감경중추대외주수술창상적응격,개선술후피로.
Objective To investigate the effects of fish oil on postoperative fatigue (POF) and the action mechansims.Methods Twenty-four SD rats were randomly divided into control group ( group C,n =8 ),surgery group (group S,n =8 ) and fish oil treatment group (group F,n =8 ).Postoperative behaviours were observed.Serum levels of interleukin (IL) -1-β,IL-6,tumor necrosis factor (TNF) -α,superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) were determined.Histological changes and ultrastructural changes of neurons in the hippocampus were examined with Hematoxylin and Eosin (HE) staining,and under an electron microscope.The central levels of 5-hydroxytryptamine (5-HT),dopamine (DA) and norepinephrine (NE) were measured by high-performance liquid chromatography-fluorescence.Results Histological and ultrastructural lesions in group F were milder than in group S.The contents of neurotransmitters were different.The levels of IL-1-β,IL-6 and TNF-α in group S [ (60.02 ± 11.08 ),(50.13 ±4.45),(41.08 ±2.17) ng/L]and group F [(45.48 ±13.51),(42.08 ±4.47),(38.76 ±2.41) ng/L]were higher than those in group C (P<0.01),and those in group F were lower than in group S ( P < 0.01 ) ; The levels of SOD and GSH-PX in group F were higher than in group S (P < 0.01 ).The results for Morris water maze in group F were superior to those in group S ( P < 0.05 ).Conclusion Fish oil could effectively ameliorate the postoperative inflammatory reaction,reduce damage to antioxidant defense system,alleviate the central stress,and improve POF.
