目的观察吗啡对人胃癌MGC-803细胞Survivin和Caspase-9基因表达的影响.方法胃癌MGC-803细胞分为两组:对照组和吗啡组.对照组不加任何药物,吗啡组将吗啡加入细胞培养液中使吗啡终质量浓度为0.1 μmol/L.孵育24h后应用流式细胞仪检测细胞凋亡,并应用半定量逆转录-聚合酶链反应(RT-PCR)和Western blot技术检测胃癌MGC-803细胞中Survivin和Caspase-9基因mRNA和蛋白的表达.结果吗啡组胃癌细胞凋亡率为(17.20±2.95)％,明显高于对照组的凋亡率(11.40±2.86)％(P＜0.05)；RT-PCR和Western blot结果显示,吗啡组胃癌细胞中Survivin mRNA和蛋白表达降低,而Caspase-9 mRNA和蛋白表达增高(P＜0.05).结论 0.1μmol/L吗啡通过抑制Survivin的表达、增强Caspase-9的活性而促进胃癌细胞的凋亡.

Objective To explore the effects of morphine on the expression of Survivin and Caspase-9 in human gastric carcinoma MGC-803 cells.Methods MGC-803 cells were divided into two groups.The cells in the control group were incubated with no drugs,while the cells in the morphine group were incubated with 0.1 μmol/L morphine.The apoptosis was assessed by flow cytometry after MGC-803 cells were incubated with morphine for 24 h.The expression of Survivin and Caspase-9 mRNA and protein was detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blotting respectively.Results The apoptosis rate of MGC-803 cells in the morphine group was higher than that in the control group (P ＜ 0.05 ).Survivin mRNA and protein expression was downregulated,while Caspase-9 mRNA and protein expression was upregulated in the morphine group (P＜0.05).Conclusion 0.1 μnol/L morphine may promote apoptosis of gastric cancer cells by Survivin downregulation and Caspase-9 upregulation in vitro.
