A method for simultaneous measurement of Acetylcholinesterase (AchE)and total cholinesterase(TchE)in rectal mucosa is described. In this procedure DFP(diisopropylfluorophosphate) is used as selective pseudocholinesterase inhibitor at a final concentration of 2×10~(-7) mol/L. We measured the rectal mucosal AchE and TchE activity of 62 children examined. Among them 24 cases suffered from Hirsch. sprung's disease, the remaining 38 were selected as a control group. The former had significantly higher AchE and TchE activity than the control, the mean AchE activity was 112.89±29.07 and 31.2±11.55×10~(-8) mol, gm~(-1) wet tissue, min~(-1), 37℃ respectively (p<0.001),the mean TchE activity was 148.55±81.99 and 58.07±18.7×10~(-8) mol,gm~(-1) wet tissue,min-1,37℃ respectively (p<0.001). The affected cut-off points of AchE and TchE were 2.5 S.D. Above normal mean similarly, approximates to 60 and 100×10~(-8) mol respectively. The diagnostic reliability of AchE was 100%and TchE was 95%. These experimental results show that the measurement of AchE and TchE activity in, rectal mucosa is a simple and valuable supplementary index in the diagnosis of Hirschsprung's disease.
