目的:观察苦参碱(Matrine,Mat)对体外人淋巴瘤Raij细胞增殖及细胞凋亡的影响,探讨 Mat 诱导 Raji 细胞凋亡的作用机制.方法:四甲基偶氮唑蓝(MTT)比色法观察Mat对Raji细胞增殖的影响;流式细胞术检测凋亡率;免疫细胞化学技术检测 Mat对Raji 细胞株 Bcl-2 蛋白表达的影响;RT-PCR法检测Bcl-2 mRNA表达.结果:不同浓度的 Mat 对 Raji 细胞均有一定的增殖抑制作用,且呈量效和时效关系;Mat能明显诱导Raji细胞凋亡;免疫细胞化学结果显示 Bcl-2 在实验组中的表达显著低于对照组.RT-PCR 显示 Bcl-2 mRNA 表达明显减弱,且随剂量增加越明显.结论:Mat 能抑制人淋巴瘤 Raji 细胞的增殖,促进细胞凋亡,其作用机制可能与下调 Bcl-2 表达有关.

Objective:To observe the effects of matrine on proliferation and apoptosis of human lymphoma cell line Raji in vitro,and to explore its mechanism.Methods:The proliferation inhibition rate of Raji cells was assayed by MTT analysis.Cell apoptosis rate was evaluated by flow cytometry.The expression level of Bcl-2 protein was evaluated by streptavidin-biotin-peroxidase method.The Bcl-2 mRNA expression of Raji cells treated with matrine was assayed by RT-PCR.Results:The matrine of different concentrations all had cytotoxic effects on Raji cells.in a dose-and time-dependent manner.The apoptosis induced by matrine was confirmed in Raji cells.Immunocytochemistry assessment showed the expression of Bcl-2 was significantly down-regulated after treated with matrine.The Bcl-2 mRNA expression in Raji cells was decreased after treated by matrine,with a concentration-effect relationship.Conclusion:Matrine can inhibit the proliferation of Raji cells significantly.By down-regulating the expression of Bcl-2 and inducing the apoptosis.
