目的:研究特异性CTL杀伤的效应与HLA型别之间的关系。方法:用已知型别的EB病毒转化的B淋巴母样细胞(Epstein-Barr-transformedB lymphoblastoid cell line,EBV-LCL)与同种异体外周血单个核细胞(PBMC)共培养,激活同种异体抗原特异性细胞毒性T细胞(cytotoxicity T cell,CTL),然后利用同位素释放法观察CTL对HLA-I类表型不同的EBV-LCL的杀伤活性。结果:用EBV-LCL-1刺激自身PBMC-1所诱导的CTL-1a,只能杀伤EBV-LCL-1,而不能杀伤HLA-I类表型不同的EBV-LCL-2;但用EBV-LCL-2刺激PBMC-1所诱导的CTL-1b,却能有效杀伤HLA-I类表型不同的EBV-LCL-2。结论:①MHC表型不同的免疫细胞之间可以发生相互作用;②TCR既不单独识别靶细胞表面的抗原肽,也不直接识别靶细胞表面的MHC分子(MHC特异性抗原决定簇),而是识别MHC-抗原肽复合物的表面综合信息,后者可能是由MHC-抗原肽复合物表面的空间构象、电荷性质及其分布等信息所构成;③所谓CTL的特异性杀伤作用,是MHC-抗原肽复合物表面信息激活该信息特异性T细胞克隆的结果。

목적:연구특이성CTL살상적효응여HLA형별지간적관계。방법:용이지형별적EB병독전화적B림파모양세포(Epstein-Barr-transformedB lymphoblastoid cell line,EBV-LCL)여동충이체외주혈단개핵세포(PBMC)공배양,격활동충이체항원특이성세포독성T세포(cytotoxicity T cell,CTL),연후이용동위소석방법관찰CTL대HLA-I류표형불동적EBV-LCL적살상활성。결과:용EBV-LCL-1자격자신PBMC-1소유도적CTL-1a,지능살상EBV-LCL-1,이불능살상HLA-I류표형불동적EBV-LCL-2;단용EBV-LCL-2자격PBMC-1소유도적CTL-1b,각능유효살상HLA-I류표형불동적EBV-LCL-2。결론:①MHC표형불동적면역세포지간가이발생상호작용;②TCR기불단독식별파세포표면적항원태,야불직접식별파세포표면적MHC분자(MHC특이성항원결정족),이시식별MHC-항원태복합물적표면종합신식,후자가능시유MHC-항원태복합물표면적공간구상、전하성질급기분포등신식소구성;③소위CTL적특이성살상작용,시MHC-항원태복합물표면신식격활해신식특이성T세포극륭적결과。
Objective:Investigate the relationship between C TL specificcytotoxicity and HLA allotype.Methods:The Epst ein-Barr-virus-transformed B lymphoblastoid cell lines(EBV-LCLs) which have different class-I HLA allotype were used as stimulators to stimulate the periph eral blood mononuclear cells (PBMC) proliferating into cytotoxic T lymphocytes(C TL),then the specific CTL was used to test cytotoxicity with different targets ( EBV-LCL mentioned above).Results:When the autologous EBV -LCL (donor 1) was used as stimulator,the CTL-1a obtained from the mixed lymph o cyte culture had cytotoxicity only targeting the EBV-LCL of donor 1,no cytotox icity targeting the EBV-LCL of donor 2.But when the allogeneous EBV-LCL (donor 2,has completely eifferent HLA-I type from donor 1)was used as stimulator,the C TL-1b showed efficient cytotoxicity to the EBV-LCL of donor 2,but very low cytotoxicity to the EBV-LCL of donor 1. Conclusion:①Immune cells with different MHC allotypes c an interact each other.②The TCRs recognize neither the antigen peptide,nor the MHC allotype determinant (MHC specific antigen determinant);what they recognize is the several information of the MHC-peptide complex surface comparised of th e conformation,the nature and disturbution of the surface charge of the MHC-pep t ide complex.③The so-called restrictive cytotoxicity of CTL results from MHC- peptide complex specifically activates the corresponding CD8+ T cell clone.
