[目的]对日本落叶松体细胞胚胎发生进行系统研究.[方法]以日本落叶松5个品种的未成熟合子胚为外植体,研究基本培养基、品种基因型、杂交系和纯系品种对日本落叶松愈伤组织诱导的影响,以及ABA对体细胞胚成熟及植株再生的影响.[结果]通过对日本落叶松胚性愈伤组织的诱导和体细胞胚的成熟培养获得再生植株.2种基本培养基中,DCR对胚性愈伤组织的诱导频率最高,达54.0%;落叶松品种之间的愈伤组织诱导率有差异,以杂交品种愈伤组织的诱导率最高,达71.4%,优于纯系的落叶松品种(27.3%~50.0%);在体细胞胚的成熟培养过程中,ABA可严重影响体细胞胚的发育,并影响到其以后的萌发能力和植株再生.[结论]杂交品系更有利于胚性愈伤组织诱导,ABA 对日本落叶松体细胞胚胎发育起重要作用.
[目的]對日本落葉鬆體細胞胚胎髮生進行繫統研究.[方法]以日本落葉鬆5箇品種的未成熟閤子胚為外植體,研究基本培養基、品種基因型、雜交繫和純繫品種對日本落葉鬆愈傷組織誘導的影響,以及ABA對體細胞胚成熟及植株再生的影響.[結果]通過對日本落葉鬆胚性愈傷組織的誘導和體細胞胚的成熟培養穫得再生植株.2種基本培養基中,DCR對胚性愈傷組織的誘導頻率最高,達54.0%;落葉鬆品種之間的愈傷組織誘導率有差異,以雜交品種愈傷組織的誘導率最高,達71.4%,優于純繫的落葉鬆品種(27.3%~50.0%);在體細胞胚的成熟培養過程中,ABA可嚴重影響體細胞胚的髮育,併影響到其以後的萌髮能力和植株再生.[結論]雜交品繫更有利于胚性愈傷組織誘導,ABA 對日本落葉鬆體細胞胚胎髮育起重要作用.

[Objective] The aim was to study the somatic embryogenesis of Larix leptolepis systematically. [Method] With premature zygotic embryos of 5 L. leptolepis cultivars as explants, the effects of basic medium, cultivar genotypes, hybrid and pure lines cultivars on the callus induction of L. leptolepis and the effects of ABA on somatic maturation and plant regeneration were studied. [Result] The regenerated plants were obtained through embryogenic callus induction of L. leptolepis and maturation culture of somatic embryo. In the 2 basic media, the induction frequency of DCR on embryogenic calli was highest, being up to 54.0%;the induction frequencies of callus had difference among L. leptolepis cultivars and that of hybrid cultivars was highest, being up to 71.4% and superior to that of pure line L. leptolepis cultivars (The difference is 27.3%-50.0%.);in the maturation culture process of somatic embryos, ABA could affect the development of somatic embryos strongly and affect their subsequent germination ability and plant regeneration. [Conclusion] The hybrid lines were more favorable to the induction of embryogenic calli and ABA played important role on the development of somatic embryogenesis of L. leptolepis.
