目的 通过测定不同时期糖尿病大鼠肾小球内血管内皮生长因子(VEGF)的表达,探讨其在糖尿病肾病的发生发展中所起的作用,并应用血管紧张素受体拮抗剂类药物缬沙坦,观察其对肾小球内VEGF表达的影响,了解其肾保护机制.方法 应用链脲佐菌素(STZ)诱导糖尿病大鼠,缬沙坦灌胃,分别在成模后2、12周时处死,免疫组化法检测肾小球内VEGF的表达.结果 糖尿病大鼠2、12周时,血糖、内生肌酐清除率、24h尿蛋白量、肾质量/体质量比均明显高于同时期正常组大鼠(P<0.01).糖尿病大鼠2、12周时肾小球内VEGF表达均明显增加(P<0.01),12周时缬沙坦组大鼠肾小球中VEGF表达强度降低,差异有统计学意义(P<0.01).结论 糖尿病早期肾小球内VEGF表达增加,可能参与早期肾小球高滤过机制.糖尿病发展到一定阶段,肾小球内VEGF表达仍高于正常,可能参与随后肾小球逐渐硬化的机制.缬沙坦可明显减少肾小球中VEGF的表达.
目的 通過測定不同時期糖尿病大鼠腎小毬內血管內皮生長因子(VEGF)的錶達,探討其在糖尿病腎病的髮生髮展中所起的作用,併應用血管緊張素受體拮抗劑類藥物纈沙坦,觀察其對腎小毬內VEGF錶達的影響,瞭解其腎保護機製.方法 應用鏈脲佐菌素(STZ)誘導糖尿病大鼠,纈沙坦灌胃,分彆在成模後2、12週時處死,免疫組化法檢測腎小毬內VEGF的錶達.結果 糖尿病大鼠2、12週時,血糖、內生肌酐清除率、24h尿蛋白量、腎質量/體質量比均明顯高于同時期正常組大鼠(P<0.01).糖尿病大鼠2、12週時腎小毬內VEGF錶達均明顯增加(P<0.01),12週時纈沙坦組大鼠腎小毬中VEGF錶達彊度降低,差異有統計學意義(P<0.01).結論 糖尿病早期腎小毬內VEGF錶達增加,可能參與早期腎小毬高濾過機製.糖尿病髮展到一定階段,腎小毬內VEGF錶達仍高于正常,可能參與隨後腎小毬逐漸硬化的機製.纈沙坦可明顯減少腎小毬中VEGF的錶達.
목적 통과측정불동시기당뇨병대서신소구내혈관내피생장인자(VEGF)적표체,탐토기재당뇨병신병적발생발전중소기적작용,병응용혈관긴장소수체길항제류약물힐사탄,관찰기대신소구내VEGF표체적영향,료해기신보호궤제.방법 응용련뇨좌균소(STZ)유도당뇨병대서,힐사탄관위,분별재성모후2、12주시처사,면역조화법검측신소구내VEGF적표체.결과 당뇨병대서2、12주시,혈당、내생기항청제솔、24h뇨단백량、신질량/체질량비균명현고우동시기정상조대서(P<0.01).당뇨병대서2、12주시신소구내VEGF표체균명현증가(P<0.01),12주시힐사탄조대서신소구중VEGF표체강도강저,차이유통계학의의(P<0.01).결론 당뇨병조기신소구내VEGF표체증가,가능삼여조기신소구고려과궤제.당뇨병발전도일정계단,신소구내VEGF표체잉고우정상,가능삼여수후신소구축점경화적궤제.힐사탄가명현감소신소구중VEGF적표체.
Objective To detect the levels of VEGF in renal cortex of diabetic nephropathy (DN) rats in different times,so as to investigate the effects of VEGF on the development of diabetic nephropathy.Meanwhile valsartan was used in the DN rat model to investigate the mechanisms of its renal protective effects on DN by observing its influence on VEGF.Methods Valsartan was perfused into the stomach of the diabetic rats induced by streptozotocin (STZ).Rats were killed after 2 weeks and 12weeks.The expression of VEGF in glomerulus was assessed by immunohistochemistry.Results Blood glucose,creatinine clearance(Ccr),24 h urinary albumin,kidney/body weight rate were significantly higher in diabetic group than that in normal group at 2nd and 12nd weeks (P < 0.01 ).There were significant upregulation of VEGF expression in glomerulus of diabetic rats at 2nd or 12nd week (P <0.01 ).At the end of 12 weeks,VEGF expression in glomerulus of rats with valsartan perfused into stomach was significantly weaker (P < 0.01 ).Conclusions At early stage of diabetic nephropathy,the expression of VEGF in kidney ascended,which was involved in the glomerulo - hyperfiltration.At late stage,it still ascended,which may be involved in the development of glomerulo - sclerosis.Valsartan can significantlv reduce the expression of VEGF in kidney.
