目的 观察RNA干扰(RNAi)对肝癌细胞株Hepal-6中乙酰肝素酶(HPSE)表达的抑制作用.方法 设计针对小鼠肝癌细胞Hepal-6 HPSE mRNA的小干扰RNA(siRNA),以阳离子脂质体LipofectamineTM 2000包埋后转染该细胞株,转染48 h后,采用逆转录-聚合酶链反应(RT-PCR)和Western blot方法检测Hepal-6细胞HPSE mRNA及蛋白质表达的变化.结果 siRNA各组与正常对照组和阴性对照组比较,HPSE mRNA和蛋白质的表达均受明显抑制(P<0.01),并且呈剂量依赖性.结论 HPSE siRNA可显著抑制小鼠肝癌细胞Hepal-6中HPSE mRNA和蛋白质的表达.

목적 관찰RNA간우(RNAi)대간암세포주Hepal-6중을선간소매(HPSE)표체적억제작용.방법 설계침대소서간암세포Hepal-6 HPSE mRNA적소간우RNA(siRNA),이양리자지질체LipofectamineTM 2000포매후전염해세포주,전염48 h후,채용역전록-취합매련반응(RT-PCR)화Western blot방법검측Hepal-6세포HPSE mRNA급단백질표체적변화.결과 siRNA각조여정상대조조화음성대조조비교,HPSE mRNA화단백질적표체균수명현억제(P<0.01),병차정제량의뢰성.결론 HPSE siRNA가현저억제소서간암세포Hepal-6중HPSE mRNA화단백질적표체.
Objective To evaluate the inhibitory effect of Heparanase (HPSE) siRNA on the ex-pression of HPSE mRNA and protein of mouse Hepal-6 hepatecarcinoma cell lines. Methods The siR-NAs which aimed at HPSE mRNA of Hepal-6,were designed and synthesized. Using cation lipofectamine 2000 embeding techniques, the siRNAs were transfeeted into Hepa1-6 cell lines for 48 hours, and then the expression of HPSE mRNA and protein were detected by RT-PCR and Western blot. Results Compared with normal and negtive contral group, HPSE mRNA and protein expression of mouse Hepal-6 cell lines in the siRNA groups were obviously decreased respectively (P < 0.01) as the siRNA' s concentration in-creased. Conclusion HPSE siRNA has remarkable inhibitory effect on the expression of HPSE mRNA and protein in mouse Hepal-6 cell lines.
