目的 探讨脑红蛋白(Ngb)mRNA在深低温停循环(DHCA)下的表达及意义.方法 建立DHCA犬模型,分别于体外循环即刻、停循环即刻、停循环60min、复温再灌注45 min,取实验犬脑皮质迅速冻存,待实验结束后统一行实时荧光PCR定量检测Ngb mRNA的表达.结果 体外循环后,特别是降温后Ngb,mRNA的拷贝均升高,至停循环60min达最高值(5.7±2.2)×108拷贝数/μl,复温再灌注后轻微下降.不同时间点Ngb mRNA的拷贝浓度间差异有统计学意义(F=8.565,P=0.1703).停循环60min时以及复温再灌注45 min后Ngb mRNA的拷贝数均显著高于体外循环即刻时(P<0.048).结论 DHCA中缺氧可诱导Ngb mRNA的表达,随着DHCA时间的延长,Ngb mRNA表达升高,复灌后表达下降.说明缺氧是决定Ngb mRNA表达高低的主要因素.

Objective To investigate the value of the neuroglobin(Ngb) related to brain injury was explored in the diagnosis, therapy and prognostic assessment of brain injury due to DHCA. Methods DHCA model was established in ten healthy adult dogs. The cerebral cortex samples were obtained and frozen for real-time fluorescence quantitative PCR (RT-PCR) for Ngb mRNA ment after extractorporeal circulation establishment, 0 and 60 minutes after circulatory arrest, and 45 minutes of reperfusion. Results The establishment of extracorporeal circulation resulted in increased Ngb mRNA expression, which was especially obvious after temperature reduction, and at 60 min after DHCA its expression reached the peak level, [±S= (5.7±2.2) ×108 copy number/μg] followed by slight decrease after body temperature recovery. The concentraion of Ngb mRNA showed significant differences at different time points( F =8.565, P = 0.003). Ngb mRNA level at 60 minutes of DHCA and at 45 minutes of reperfusion was significantly higher than that measured before extracorporeal circulatio establishment and at the start of DHCA(P<0.048). Conclusion Anoxia in DHCA cam induce the Ngb mRNA expression. As DHCA was prolonged, Ngb mRNA level kept increasing but the increment was of gradually lowered magnitudes.
