哺乳动物雷帕霉素靶蛋白(mTOR)是PI3K/Akt/mTOR信号通路下游的重要效应蛋白,其底物主要控制与细胞生长和增殖密切相关的蛋白质的合成.mTOR在多种恶性肿瘤中表达异常,是肿瘤靶向治疗的新靶点.临床前研究和Ⅰ、Ⅱ期临床实验表明,以雷帕霉素及其衍生物为代表的mTOR抑制剂和其它传统抗肿瘤药物的联合应用取得了良好的的抗肿瘤效果,为肿瘤的靶向治疗提供了新的方向.

The mammalian target of rapamycin ( mTOR) is an important effector of the PDK/Akt/ MTOR pathway, which is centrally involved in growth regulation, proliferation control, and cancer cell metabolism. Over-expressions of mTOR can be found in many malignancies, which make mTOR a potential target for cancer therapy. The mTOR inhibitors (rapamycin and its analogues) in combination with traditional anti-tumor drugs have shown reasonable effect in cancer treatment both in pre-clinical experiments and in some stage Ⅰ and Ⅱ clinical trials. The studies provide a new aspect for the targeted treatment of tumors.
