目的 观察胶质瘤患者肿瘤组织中人类白细胞抗原(HLA)Ⅰ类抗原及抗原转运相关蛋白(TAP)1、TAP2的表达,探讨其与肿瘤病理级别的关系.方法 收集41例经病理确诊的胶质瘤患者肿瘤组织标本,通过免疫组织化学的方法,观察不同病理级别肿瘤标本中HLA Ⅰ类抗原及抗原加工递呈(APM)分子的表达水平,探讨HLA Ⅰ类抗原和APM分子表达与胶质瘤患者病理级别之间的关系.结果 HLA Ⅰ类抗原丢失在各级别胶质瘤组织中分别为11% (1/9)、25% (3/12)、30%(3/10)及40% (4/10).TAP1和TAP2蛋白水平均与胶质瘤分级明显相关(P<0.05),表达水平随胶质瘤分级增加表达下降.此外,TAP1表达水平与TAP2明显相关(P<0.05).结论 胶质瘤组织中HLA Ⅰ抗原和部分APM分子蛋白水平,随胶质瘤级别增加而降低;这可能是导致部分T细胞免疫治疗失败的原因之一.

목적 관찰효질류환자종류조직중인류백세포항원(HLA)Ⅰ류항원급항원전운상관단백(TAP)1、TAP2적표체,탐토기여종류병리급별적관계.방법 수집41례경병리학진적효질류환자종류조직표본,통과면역조직화학적방법,관찰불동병리급별종류표본중HLA Ⅰ류항원급항원가공체정(APM)분자적표체수평,탐토HLA Ⅰ류항원화APM분자표체여효질류환자병리급별지간적관계.결과 HLA Ⅰ류항원주실재각급별효질류조직중분별위11% (1/9)、25% (3/12)、30%(3/10)급40% (4/10).TAP1화TAP2단백수평균여효질류분급명현상관(P<0.05),표체수평수효질류분급증가표체하강.차외,TAP1표체수평여TAP2명현상관(P<0.05).결론 효질류조직중HLA Ⅰ항원화부분APM분자단백수평,수효질류급별증가이강저;저가능시도치부분T세포면역치료실패적원인지일.
Objective To observe the expression of human leukocyte antigen Ⅰ ( HLA Ⅰ) and antigen processing machinery (APM) components,and their correlation with pathological grading.Methods Forty-one tumor sections from forty-one glioma patients were examined by using immunohistochemistry.The expression of HLA Ⅰ antigen and APM components was detected,and their correlation with the pathological grading was analyzed.Results Frequency of HLA Ⅰ antigen loss in dedicated groups was 11%,25%,30% and 40%,respectively.There was a positive correlation between the expression levels of transporter associated with antigen processing (TAP) 1 and TAP2 with glioma pathological grading (P < 0.05 ).Additionally,there was a positive correlation between TAP1 and TAP2 level (P < 0.05 ).Conclusion The level of HLA Ⅰ antigen and most of APM components expression is decreased with the increase in tumor pathological grade,which may explain the phenomenon that glioma cells frequently escape from immunological surveillance or immunotherapy.
