目的 探讨食管鳞癌患者外周血循环肿瘤细胞数量与食管癌病理生理特征之间的关系.方法 采取负性筛选策略,红细胞裂解液去除食管鳞癌外周血样本中的红细胞,Miltenyi免疫磁珠去除白细胞,富集上皮来源的稀有细胞,免疫荧光染色技术鉴定循环肿瘤细胞.结果 食管鳞癌患者外周血循环肿瘤细胞的数量与患者性别、年龄、肿瘤位置均无显著相关性,与细胞分化程度、原发肿瘤侵犯程度、淋巴结转移状态、P-TNM分期有显著相关,相关系数分别为0.504、0.507、0.412、0.374.结论 食管鳞癌患者外周血循环肿瘤细胞的数量可以反映肿瘤的进展程度,可作为判断食管鳞癌生物学行为恶化的指标.

Objective To explore the relationship between circulating tumor cells(CTCs) in the peripheral blood of patients with esophageal squamous cell carcinoma(ESCC) and the physiopathological characteristics of esophageal neoplasms.Methods Using negative selection system,we depleted red blood cells(RBCs) in red blood cell lysis buffer,depleted white blood cells (WBCs) with Miltenyi magnetic beads and enriched the rare cells from ESCC patients'peripheral blood.Immunofluorence staining (IF) was adopted to identify CTCs.Results Circulating tumor cells in the peripheral blood of patients with esophageal squamous cell carcinoma was closely related to cell differentiation grade,the invasion of primary cancer,lymph node status,P-TNM stages,and was rarely related to the sex,age or the location of tumor.Conclusion The results suggest that circulating tumor cells in the peripheral blood of patients with esophageal squamous cell carcinoma may express the development of esophageal cancer and may be served as a tumor marker to evaluate the biological behavior of esophageal cancer.
