目的研究DT对PC-3细胞livin基因表达的影响.方法予10～7mol/L浓度DT体外作用PC-3细胞48h,RT-PCR及westerm-blot检测处理组与对照组PC-3细胞livin基因mRNA及蛋白表达水平变化.结果处理组与对照组PC-3细胞livin基因mRNA及蛋白表达水平分别为(55.17 ± 0.87)%、(8.23±0.68)%;(87.32±9.13)%、(51.12±2.01)%;经t检验处理组与对照组差别有统计学意义.结论 DT处理PC-3细胞后livin基因表达增强,livin基因可能在PC-3细胞对DT化疗耐药中起重要作用.

Objectives To study the effect of DT on the expression of livin gene of PC - 3 cell. Methods The PC - 3 cell was treated with 10 ～ 7mol/L DT in vitro for 48h, then the expression level of livin gone mRNA and protein was tested by RT - PCR and western - blot. Results The livin mRNA and protein expression of treated group and control group PC - 3 cell was respectively (55.17 ± 0.87) %, (8.23 ± 0.68) % ; (87.32 ± 9.13) %, (51.12 ± 2.01) % ; the differences between the treated group and control group have statistical significance by t test. Conclusions The livin gane expression of PC - 3 cells rise after treated with DT, livin gene may play an important role in drug resistance in the PC - 3 cells to DT chemotherapy.
