目的探讨细胞周期调控抑制因子 p27kip1蛋白和周期蛋白 E(cyclinE)在大肠腺癌发生发展中的作用.方法采用免疫组化技术 SP法,检测 p27kip1和 cyclinE蛋白在 37例大肠腺癌和 26例正常大肠黏膜组织中的表达.结果 p27kip1蛋白阳性表达率在大肠腺癌组织中为 45.9% , 显著低于正常大肠黏膜组织 (χ 2=4.590, P< 0.05),并与大肠腺癌组织分化程度和淋巴结转移均有关 (χ 2=9.653, χ 2=6.838, P< 0.05); cyclinE蛋白阳性表达率在大肠腺癌组织中为 64.9%,显著高于正常大肠黏膜组织 (χ 2=4.285, P< 0.05) , 并与大肠腺癌组织分化程度和淋巴结转移均有关 (χ 2=8.397,χ 2=5.261, P< 0.05); p27kip1蛋白阳性表达组 cyclinE蛋白阳性表达率低于 p27kip1蛋白阴性表达组,但两者无相关( r=- 0.1133, P >0.05).结论 p27kip1蛋白的低表达和 /或 cyclinE蛋白的高表达在大肠腺癌发生中发挥重要作用,并可能有助于预测其恶性程度及淋巴结转移,可能对大肠腺癌的治疗和生活质量的改善有指导作用.

목적탐토세포주기조공억제인자 p27kip1단백화주기단백 E(cyclinE)재대장선암발생발전중적작용.방법채용면역조화기술 SP법,검측 p27kip1화 cyclinE단백재 37례대장선암화 26례정상대장점막조직중적표체.결과 p27kip1단백양성표체솔재대장선암조직중위 45.9% , 현저저우정상대장점막조직 (χ 2=4.590, P< 0.05),병여대장선암조직분화정도화림파결전이균유관 (χ 2=9.653, χ 2=6.838, P< 0.05); cyclinE단백양성표체솔재대장선암조직중위 64.9%,현저고우정상대장점막조직 (χ 2=4.285, P< 0.05) , 병여대장선암조직분화정도화림파결전이균유관 (χ 2=8.397,χ 2=5.261, P< 0.05); p27kip1단백양성표체조 cyclinE단백양성표체솔저우 p27kip1단백음성표체조,단량자무상관( r=- 0.1133, P >0.05).결론 p27kip1단백적저표체화 /혹 cyclinE단백적고표체재대장선암발생중발휘중요작용,병가능유조우예측기악성정도급림파결전이,가능대대장선암적치료화생활질량적개선유지도작용.
Aim The relationships between p27kip1 protein as negative factor of cell cycle regulation and colorectal adenocarcinoma have been seldomly reported in China.This article aims to investigate the effects of inhibiting factor of cell cycle regulation-p27kip1 and cyclinE proteins in the genesis and progression of human colorectal adenocarcinoma.Methods The expressions of p27kip1 and cyclinE proteins in tumor tissues of 37 patients with colorectal adenocarcinoma and in normal colorectal mucosal tissues of 26 subjects were detected by SP immunohistochemical technique.Results p27kip1 protein positive-expression rate in tumor tissues of colorectal adenocarcinoma was 45.9% , which was lower than that in normal colorectal mucosal tissues (χ 2=4.590,P< 0.05), p27kip1 protein positive-expression correlated significantly with tumor cell differentiation and lymph node metastasis (χ 2=9.653;χ 2=6.838,P< 0.05); cyclinE positive-expression rate in tumor tissues was 64.9%, which was higher than that in normal colorectal mucosal tissues (χ 2=4.285,P< 0.05); cyclinE positive-expression also correlated significantly with tumor cell differentiation and lymph node metastasis (χ 2=8.397;χ 2=5.261,P< 0.05); cyclinE protein positive-expression rate in the tumor tissues of p27kip1 protein positive-expression group was lower than that in p27kip1 protein negative-expression group, there were no significantly correlation between the two groups ( r =- 0.1133, P >0.05).Conclusion The decreased expression of p27kip1 protein and/or over- expression of cyclinE protein may play an important role in the genesis of colorectal adenocarcinoma and might be helpful to the determination of malignant degree and lymph node metastasis as well as the treatment and living quality of the diseases.
