Type 1 diabetes IL-4 NOD mice Imunotherapy
目的探讨IL-4对NOD鼠1型糖尿病发病率、胰岛炎的影响及其机制。方法采用人类1型糖尿病动物模型NOD(nonobe se diabetic)鼠,IL-4处理后检测血糖、尿糖及糖尿病发病率,HE染色观察胰岛炎程度,流式细胞仪测定周围及中枢淋巴细胞亚群的变化。结果 IL-4处理组糖尿病发病率为12.50%,明显低于对照组(62.50%)(P<0.05),且胰岛炎的严重程度亦低于对照组;处理组周围单个核细胞CD+4/CD+8及IL-2R+细胞百分比增高,胸腺CD+4 CD+8细胞百分比增高而CD+4 CD+8细胞百分比下降(P<0.01) 。结论 IL-4有降低NOD鼠1型糖尿病发病率及胰岛炎严重程度的作用;这种作用可能与IL-4影响了淋巴细胞亚群的分布从而纠正了免疫调节失衡有关。

목적탐토IL-4대NOD서1형당뇨병발병솔、이도염적영향급기궤제。방법채용인류1형당뇨병동물모형NOD(nonobe se diabetic)서,IL-4처리후검측혈당、뇨당급당뇨병발병솔,HE염색관찰이도염정도,류식세포의측정주위급중추림파세포아군적변화。결과 IL-4처리조당뇨병발병솔위12.50%,명현저우대조조(62.50%)(P<0.05),차이도염적엄중정도역저우대조조;처리조주위단개핵세포CD+4/CD+8급IL-2R+세포백분비증고,흉선CD+4 CD+8세포백분비증고이CD+4 CD+8세포백분비하강(P<0.01) 。결론 IL-4유강저NOD서1형당뇨병발병솔급이도염엄중정도적작용;저충작용가능여IL-4영향료림파세포아군적분포종이규정료면역조절실형유관。
Objective To study the effectiveness of IL-4 on type 1 diabetes in NOD mice and it’s possible mechanism.Methods 4-week-old NOD mice,as a kind of type 1 diabetes model,were treated with IL-4 to investigate the incidence of insulitis,and lymphocyte subsets were determined by FACS.Results The incidence of diabetes in the IL-4 group was 12.50% which was obviously less than 62.50% in the control group ( P <0.05),and the degree of insulitis in the IL-4 group was also less than that in the control group.The lymphocyte subset in the IL-4 treated group showed higher CD + 4/CD + 8 cells and percentage of IL-2R + cells in spleen,and higher proportion of CD + 4/CD + 8 but decreased CD + 4/CD + 8 cells in thymus.Conclusion IL-4 can prevent NOD mice from developing type 1 diabetes,which may be related to the fact that IL-4 corrected the imbalance of immunoregulatory in NOD mice.
