目的 通过弹性蛋白主动免疫及熏烟制造豚鼠肺气肿模型,观察各模型中肺组织和心肌组织基质金属蛋白酶9(MMP-9)、MMP - 12及干扰素介导蛋白10(IP-10)的表达情况及组织破坏情况,探究MMP-9 、MMP- 12及IP- 10在自体免疫反应所致肺气肿形成过程中的作用.方法 将24只雄性豚鼠随机分为3组:主动免疫组、熏烟组、对照组.主动免疫组于皮下多点注射弹性蛋白;熏烟组采用传统熏烟造模;对照组不做任何处理.造模满12周后进行肺组织及心肌组织的病理形态观察.并用免疫组织化学法观察各组组织中的MMP-9 、MMP-12及IP-10蛋白的表达情况.结果 主动免疫组及熏烟组肺组织及心肌组织中MMP -9、MMP-12及1P-10均较对照组增多(P<0.05).主动免疫组肺组织中MMP-9、MMP -12的蛋白表达与熏烟组差异无统计学意义(P>0.05).主动免疫组心肌组织中MMP-9、MMP- 12及IP -10较熏烟组明显增多(P<0.05).结论 弹性蛋白抗体自体免疫可以诱发豚鼠肺气肿的形成.MMP-9、MMP -12及IP-10在自体免疫反应所致肺气肿的发生发展中有重要的促进作用.

목적 통과탄성단백주동면역급훈연제조돈서폐기종모형,관찰각모형중폐조직화심기조직기질금속단백매9(MMP-9)、MMP - 12급간우소개도단백10(IP-10)적표체정황급조직파배정황,탐구MMP-9 、MMP- 12급IP- 10재자체면역반응소치폐기종형성과정중적작용.방법 장24지웅성돈서수궤분위3조:주동면역조、훈연조、대조조.주동면역조우피하다점주사탄성단백;훈연조채용전통훈연조모;대조조불주임하처리.조모만12주후진행폐조직급심기조직적병리형태관찰.병용면역조직화학법관찰각조조직중적MMP-9 、MMP-12급IP-10단백적표체정황.결과 주동면역조급훈연조폐조직급심기조직중MMP -9、MMP-12급1P-10균교대조조증다(P<0.05).주동면역조폐조직중MMP-9、MMP -12적단백표체여훈연조차이무통계학의의(P>0.05).주동면역조심기조직중MMP-9、MMP- 12급IP -10교훈연조명현증다(P<0.05).결론 탄성단백항체자체면역가이유발돈서폐기종적형성.MMP-9、MMP -12급IP-10재자체면역반응소치폐기종적발생발전중유중요적촉진작용.
Objective To make emphysema models by active immunization and cigarette smoking using guinea pigs,then observe the expression of MMP-9,MMP-12,IP-10 and the destruction of pulmonary tissue and myocardial tissue in each model,to explore the role of the MMP-9,MMP-12,IP-10in the emphysema which induced by active immunization.Methods The guinea pigs were randomly divided into three groups,involving active immunization group,smoking group and control group.The active immunization group used the method of subcutaneous injection of elastin at multiply points and the smoking group used the traditional method,while the control group performed nothing.To observe the morphosis of pulmonary tissue and myocardial tissue and the expression of MMP-9,MMP -12,IP-10 in the pulmonary tissue and myocardial tissue of every model were observed by immunohistochemical staining,Results The expression of MMP-9,MMP-12,IP -10 in the pulmonary tissue and myocardial tissue has both significantly increased in the active immunization group and the smoking group,compared with the control group ( P <0.05).The expression of MMP-9,MMP-12 in the pulmonary tissue in the active immunization group compared with the smoking group,and there is no obvious difference ( P >0.05).The expression of matrix MMP 9,MMP-12 in the myocardial tissue in the active immunization group,compared with the smoking group ( P < 0.05).Conclusions Emphysema model of guinea pig can be established by active immunization.MMP-9,MMP-12,IP- 10 has important positive influence on emphysema which induced by active immunization.
