Huoxue Xifeng Recipe%Photo-chemistry%VEGF mRNA%RT-PCR%Focal cerebral infarction
目的 观察活血熄风方对局灶性脑缺血模型大鼠脑梗塞体积及脑组织中血管内皮生长因子(VEGF mRNA)表达的影响.方法 Wistar雄性大鼠随机分为假手术组,模型组,活血熄风方高、中、低3个剂量组(剂量分别为20,10,5 g·kg~(-1))和尼莫地平对照组(0.02 g·kg~(-1)),采用光化学法建立局灶性脑缺血大鼠模型,造模前7d灌胃给药,造模后24 h 取材,TTC染色法测脑梗塞体积,逆转录聚合酶链式反应(RT-PCR)法测VEGF mRNA表达.结果 与模型组比较,活血熄风方各剂量组的脑梗塞体积明显减少(P<0.01),VEGF mRNA表达明显升高 (P<0.01).结论 活血熄风方可明显减少局灶性脑缺血大鼠脑梗塞体积,具有脑保护作用,其作用机制可能与增强VEGF mRNA表达有关.

목적 관찰활혈식풍방대국조성뇌결혈모형대서뇌경새체적급뇌조직중혈관내피생장인자(VEGF mRNA)표체적영향.방법 Wistar웅성대서수궤분위가수술조,모형조,활혈식풍방고、중、저3개제량조(제량분별위20,10,5 g·kg~(-1))화니막지평대조조(0.02 g·kg~(-1)),채용광화학법건립국조성뇌결혈대서모형,조모전7d관위급약,조모후24 h 취재,TTC염색법측뇌경새체적,역전록취합매련식반응(RT-PCR)법측VEGF mRNA표체.결과 여모형조비교,활혈식풍방각제량조적뇌경새체적명현감소(P<0.01),VEGF mRNA표체명현승고 (P<0.01).결론 활혈식풍방가명현감소국조성뇌결혈대서뇌경새체적,구유뇌보호작용,기작용궤제가능여증강VEGF mRNA표체유관.
Objective To observe the effects of Huoxue Xifeng Recipe (HXR) on infarction volume and the expressions of VEGF mRNA in brain tissue after focal cerebral infarction(FCI) in rats.Methods Wistar male rats were randomly divided into six groups: sham group, model group, HXR treated group(20,10,5 g·kg~(-1) as high,middle and low dose respectively) and nimodipine control group(0.02 g·kg~(-1)). The model of FCI was established based on the principle of photo-chemistry initiation of thrombosis, intragastric administration continued 7 days before modeling and draw the material after 24h.Infarction volume was detected by TTC staining technique,the expression of VEGF mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR). Results The infarction volume was decreased and the expressions of VEGF mRNA was higher in HXR trerted groups than that of FCI model rats (P<0.05 or P<0.01).Conclusion HXR can decreased infarction volume and has a protective effect on FCI rats, and the mechanism may be related to increasing the expression of VEGF mRNA.
