目的 研究不同浓度苯妥英钠对化疗耐药胶质母细胞瘤细胞系(8-MG-BA)化疗耐药逆转的影响.方法 用不同浓度苯妥英钠对8-MG-BA及H4细胞系处理后,以噻唑蓝检测其对上述2种细胞系化疗耐药逆转的影响,并与维拉帕米处理组进行对照.结果 用苯妥英钠5、10 mg/L和维拉帕米5 mg/L处理后,8-MG-BA细胞对卡莫司汀的逆转倍数分别为25.34、34.97和14.27,对替尼泊苷耐药的逆转倍数分别为13.44、21.71和11.10.结论 苯妥英钠是一种新型多药耐药逆转剂,逆转作用大于维拉帕米,且呈现剂量依赖关系.

0bjective To investigate the reversal of chemotherapy resistance of glioblastoma cell line 8-MG-BA after incubated in different concentrations of phenytoin(PHT)in vitro.Methods After adding PHT on different concentrations,the reversal of chemotherapy resistance of carmustine and VM-26 on H4 and 8-MG-BA cell line was determined by thiazolyl blue tetrazolium bromide(MTr)assay.Verapamil was applied in a control group.Results With PHT 5mg/L or 10 mg/L,verapamil(VRP)5 mg/L,the fold of reversal of carmustine was 25.34,34.97,14.27 respectively,and that of VM-26 was 13.44,21.71,11.10 respectively.Conclusion Fold of reversal increases more with the dose of PHT that with VRP.
