目的 探讨肝细胞生长因子激活因子的抑制因子(HAI-1)在前列腺癌(PCa)组织中的表达及其意义.方法 收集20例前列腺增生(BPH)和48例PCa患者的前列腺组织标本,应用逆转录-聚合酶链反应(RT-PCR)、蛋白印迹(Western blot)法分别从mRNA水平、蛋白水平检测HAI-1基因的表达.结果 HAI-1的阳性率在BPH和PCa中为85.00%、52.08% (P <0.05);HAI-1 mRNA的相对表达值分别为0.5482±0.0313、0.6718±0.0516(P <0.05);HAI-1蛋白的相对表达值分别为0.5975±0.0518、0.7039±0.0377(P<0.05);随细胞分化程度的降低、临床分期的递增,HAI-1基因的表达量下降(P<0.05),有淋巴结转移组显著低于无淋巴结转移组(P<0.05).结论 HAI-1基因的表达缺失可作为前列腺癌浸润、转移和预后的一种分子标志.

Objective To investigate the expression and clinical significance of hepatocyte growth factor activator inhibitor type 1 ( HAI-1 ) gene in the tissues of prostatic carcinoma (PCa).Methods The prostate tissue specimens from 20 patients with benign prostate hyperplasia (BPH) and 48 patients with PCa were collected.Reverse transcription polymerase chain reaction ( RT-PCR ) and Western blotting were used to detect the expression of HAI-1 in these tissues.Results The positive expression rate of HAI-1 was 85.00% and 52.08% respectively in 20 cases of BPH and 48 cases of PCa tissues (P <0.05).The relative expression values of HAI-1 mRNA were 0.5482 ±0.0313 and 0.6718 ± 0.0516 respectively in PCa and BPH ( P < 0.05 ).The relative expression values of HAI-1 protein in PCa and BPH tissues were 0.5975 ± 0.0518 and 0.7039 ± 0.0377 respectively (P < 0.05 ).With the decrease in cellular differentiation and increase in the clinical staging,the expression of HAI-1 gene was decreased ( P < 0.05 ).The expression of HAI-1 gene in lymph node metastasis group was significantly lower than that in non-lymph node metastasis group (P < 0.05 ).Conclusion The HAI-1 gene deletion can be used as an important molecular marker in the invasion and metastasis of PCa.
