目的 研究蛋氨酸胆碱对铅暴露大鼠海马CaMKⅡ和CREB mRNA和蛋白表达的影响.方法 将断乳健康SPF级雄性SD大鼠分为空白对照组、染铅组(0.4 g/L乙酸铅饮水染毒)、蛋氨酸胆碱组(1:1,400 mg/kg)、铅+低剂量蛋氨酸胆碱组和铅+高剂量蛋氨酸胆碱组.铅+低、高剂量蛋氨酸胆碱组在染铅的同时,经灌胃分别给予低、高剂量(100、400 mg/kg)蛋氨酸胆碱混合溶液(1:1),8周后取大鼠海马组织,用实时聚合酶链反应(PCR)法测定CaMKⅡ和CREB mRNA的表达水平,免疫印迹(Western blot)法测定CaMKⅡ、CREB和pCREB蛋白含量.结果 染铅组大鼠海马CaMKⅡI和CREB mRNA的表达水平(0.743±0.185,0.729±0.199)均明显低于空白对照组(0.950±0.238,0.901±0.232),CREB和pCREB蛋白表达量( 0.271±0.045,0.212±0.058)低于空白对照组(0.319±0.058,0.506±0.125),差异均有统计学意义(P<0.05);给予低、高剂量蛋氨酸胆碱后染铅大鼠海马CaMKⅡ mRNA表达水平(1.014±0.210,1.126±0.379)和CREB mRNA表达水平(1.029±0.335,0.932±0.251)均明显高于染铅组,差异有统计学意义(P<0.05),铅+高剂量蛋氨酸胆碱组CREB和pCREB蛋白表达量(0.407±0.951,0.563±0.178)明显高于染铅组,差异有统计学意义(P<0.05).结论 蛋氨酸胆碱可以拮抗铅对海马CaMKⅡ和CREB mRNA和蛋白表达的抑制作用.

목적 연구단안산담감대연폭로대서해마CaMKⅡ화CREB mRNA화단백표체적영향.방법 장단유건강SPF급웅성SD대서분위공백대조조、염연조(0.4 g/L을산연음수염독)、단안산담감조(1:1,400 mg/kg)、연+저제량단안산담감조화연+고제량단안산담감조.연+저、고제량단안산담감조재염연적동시,경관위분별급여저、고제량(100、400 mg/kg)단안산담감혼합용액(1:1),8주후취대서해마조직,용실시취합매련반응(PCR)법측정CaMKⅡ화CREB mRNA적표체수평,면역인적(Western blot)법측정CaMKⅡ、CREB화pCREB단백함량.결과 염연조대서해마CaMKⅡI화CREB mRNA적표체수평(0.743±0.185,0.729±0.199)균명현저우공백대조조(0.950±0.238,0.901±0.232),CREB화pCREB단백표체량( 0.271±0.045,0.212±0.058)저우공백대조조(0.319±0.058,0.506±0.125),차이균유통계학의의(P<0.05);급여저、고제량단안산담감후염연대서해마CaMKⅡ mRNA표체수평(1.014±0.210,1.126±0.379)화CREB mRNA표체수평(1.029±0.335,0.932±0.251)균명현고우염연조,차이유통계학의의(P<0.05),연+고제량단안산담감조CREB화pCREB단백표체량(0.407±0.951,0.563±0.178)명현고우염연조,차이유통계학의의(P<0.05).결론 단안산담감가이길항연대해마CaMKⅡ화CREB mRNA화단백표체적억제작용.
Objective To study the effects of methionine and choline on the expression levels of CaMKⅡ and CREB mRNA and proteins in hippocampus of rats exposed to lead.Methods Male SD rats were divided into five groups.( 1 ) control group,(2) group exposed to lead+2 by drinking water with 0.40 g/L lead acetate,(3) group exposed to methionine and choline ( 1:1,400 mg/kg),(4) group exposed to 0.40 g/L lead acetate plus methionine and choline (1:1,100 mg/kg),(5) group exposed to 0.40 g/L lead acetate plus methionine and choline ( 1:1,400 mg/kg).In 8 weeks after exposure,all rats were killed.Then CREB mRNA and CaMK Ⅱ mRNA expression levels in hippocampus were detected by real-time PCR,CREB and CaMK Ⅱprotein expression levels in hippocampus were measured by western blot assay.Results The expression levels (0.743±0.185 and 0.729±0.199) of CaMKⅡ mRNA and CREB mRNA in the hippocampus of lead group were significantly lower than those (0.950±0.238 and 0.901 ±0.232) of control group (P<0.05),also the expression levels (0.271 ±0.045 and 0.212±0.058) of CREB protein and pCREB protein in the hippocampus of lead group were significantly lower than those (0.319±0.058 and 0.506±0.125) of control group (P<0.05).The expression levels (1.014±0.210 and 1.126±0.379) of CaMKⅡ mRNA and the expression levels (1.029±0.335 and 0.932±0.251 ) of CREB mRNA in the hippocampus of 2 groups exposed to lead acetate plus methionine and choline were significantly higher than those of lead group (P<0.05).The expression levels (0.407±0.951 and 0.563±0.178) of CREB protein and pCREB protein in the hippocampus of group exposed to lead acetate plus 400 mg/kg methionine and choline were significantly higher than those of lead group (P<0.05).Conclusion Methionine and choline could decrease the inhibition effects of lead on the expression of CaMKⅡ and CREB mRNA orCREB and pCREB proteins in the hippocampus of rats.
