目的 观察胰岛素样生长因子(IGF)-1基因转染的脂肪间充质干细胞(ADSCs)向软骨细胞分化的效果.方法 原代培养兔ADSCs,免疫荧光法检测细胞表面抗原CD44、CIM9;脂质体介导人IGF-1基因转染兔ADSCs联合低浓度血清培养基向软骨细胞分化诱导,RT-PCR及Western blot方法检测IGF-1的表达,MMT法绘制细胞增殖曲线、甲苯胺蓝染色软骨结节、免疫组织化学检测Ⅱ型胶原的表达.结果 脂肪间充质干细胞CD44、CD109表达阳性,基因转染后细胞IGF-1表达阳性,细胞增殖速度增快,出现软骨结节,Ⅱ型胶原表达增高.结论 从脂肪组织中能够分离出增殖旺盛的ADSCs,IGF-1在ADSCs内获得稳定表达,细胞增殖能力增强,促进其向软骨细胞分化.
目的 觀察胰島素樣生長因子(IGF)-1基因轉染的脂肪間充質榦細胞(ADSCs)嚮軟骨細胞分化的效果.方法 原代培養兔ADSCs,免疫熒光法檢測細胞錶麵抗原CD44、CIM9;脂質體介導人IGF-1基因轉染兔ADSCs聯閤低濃度血清培養基嚮軟骨細胞分化誘導,RT-PCR及Western blot方法檢測IGF-1的錶達,MMT法繪製細胞增殖麯線、甲苯胺藍染色軟骨結節、免疫組織化學檢測Ⅱ型膠原的錶達.結果 脂肪間充質榦細胞CD44、CD109錶達暘性,基因轉染後細胞IGF-1錶達暘性,細胞增殖速度增快,齣現軟骨結節,Ⅱ型膠原錶達增高.結論 從脂肪組織中能夠分離齣增殖旺盛的ADSCs,IGF-1在ADSCs內穫得穩定錶達,細胞增殖能力增彊,促進其嚮軟骨細胞分化.
목적 관찰이도소양생장인자(IGF)-1기인전염적지방간충질간세포(ADSCs)향연골세포분화적효과.방법 원대배양토ADSCs,면역형광법검측세포표면항원CD44、CIM9;지질체개도인IGF-1기인전염토ADSCs연합저농도혈청배양기향연골세포분화유도,RT-PCR급Western blot방법검측IGF-1적표체,MMT법회제세포증식곡선、갑분알람염색연골결절、면역조직화학검측Ⅱ형효원적표체.결과 지방간충질간세포CD44、CD109표체양성,기인전염후세포IGF-1표체양성,세포증식속도증쾌,출현연골결절,Ⅱ형효원표체증고.결론 종지방조직중능구분리출증식왕성적ADSCs,IGF-1재ADSCs내획득은정표체,세포증식능력증강,촉진기향연골세포분화.
Objective To explore the biologic activity of adipose mesenchymal stem cells(ADSCs) and the effect of IGF-1 gene in transforming ADSCs into chondrocytes in vitro.Methods Primary culture of the ADSCs from fat tissue was done.CD44 and CD109 were detected by immunotluorescence.ADSCs were transfected with the method of stable transfection of gene IGF-1 with Liprofectamine 2000 and the low concentration blood serum culture-medium.The expression of IGF-1 was detected by the method of RT-PCR and Western blot.The proliferation of ADSCs was assayed MTT.The carthage nodus was analyzed bv toluidine blue stain.The expression of collagen Ⅱ was tested by immunofluorescence.Results ADSCs were positive for CD44 and CD109.After gene transfection,the ADSCs expressed IGF-1,the prolifcration of ADSCs was increased,cartilage nodus was seen and the expression of collagen Ⅱ up-regulated.Conclusion Exogenous IGF-1 was stably expressed in ADSCs.Tranfection of IGF-1 into ADSCs increaSed their proliferation and promote the difrerentiation into chondrocytes in vitro.
