目的:用化学发光法测定生物制剂样品中SOD活性并消除样品中天然自由基清除剂黄酮类及维生素C、E等成分的干扰。方法:分别用黄嘌呤——黄嘌呤氧化酶及邻苯三酚自氧化作为酶和非酶超氧阴离子自由基发生体系,以鲁米诺为发光剂,通过样品对发光的抑制率测定其SOD活性;并通过加热样品使SOD失活测得样品发光抑制率本底值,扣除以消除干扰。结果:用本法消除干扰后,同一样品在两种体系中测得结果有高度一致性,SOD活性分别为170 U/ml和165 U/ml;不经加热处理直接测定值为4265 U/ml和275 U/ml。结论:本法用于测定生物制剂中SOD活性,可方便有效地消除黄酮类及维生素C、E等成分的干扰。
目的:用化學髮光法測定生物製劑樣品中SOD活性併消除樣品中天然自由基清除劑黃酮類及維生素C、E等成分的榦擾。方法:分彆用黃嘌呤——黃嘌呤氧化酶及鄰苯三酚自氧化作為酶和非酶超氧陰離子自由基髮生體繫,以魯米諾為髮光劑,通過樣品對髮光的抑製率測定其SOD活性;併通過加熱樣品使SOD失活測得樣品髮光抑製率本底值,釦除以消除榦擾。結果:用本法消除榦擾後,同一樣品在兩種體繫中測得結果有高度一緻性,SOD活性分彆為170 U/ml和165 U/ml;不經加熱處理直接測定值為4265 U/ml和275 U/ml。結論:本法用于測定生物製劑中SOD活性,可方便有效地消除黃酮類及維生素C、E等成分的榦擾。
목적:용화학발광법측정생물제제양품중SOD활성병소제양품중천연자유기청제제황동류급유생소C、E등성분적간우。방법:분별용황표령——황표령양화매급린분삼분자양화작위매화비매초양음리자자유기발생체계,이로미낙위발광제,통과양품대발광적억제솔측정기SOD활성;병통과가열양품사SOD실활측득양품발광억제솔본저치,구제이소제간우。결과:용본법소제간우후,동일양품재량충체계중측득결과유고도일치성,SOD활성분별위170 U/ml화165 U/ml;불경가열처리직접측정치위4265 U/ml화275 U/ml。결론:본법용우측정생물제제중SOD활성,가방편유효지소제황동류급유생소C、E등성분적간우。
Objective To develop a Chemiluminescent assay method for detecting SOD activity in biological samples,eliminating the interference of the natural free radical cavenger in sample,such as flavonoids,Vitamin C and E.Methods SOD activity in the samples was calculated with the inhibitory rate of luminol chemiluminescence of superoxide anion generated by Xanthine-Xanthine oxidase and by autoxidation ac pyrogallol systems,respectivdy, and the interference was determined by heating the samples at 100℃ water-bath to obtain the blank of samples. Results After eliminating the interference SOD activity in the same sample measured with two systems was 170 and 165 U/ml,respectively, while the non-heating samples’were 4265 U/ml and 275 U/ml respectively.Conclusion This technique for determining SOD activity in the biological preparations can conveniently and effectirely eliminate the interference from flavonoids,vitamin C and E.
