目的 探讨肾脏20-HETE下调在高脂饮食所致高血压发病机制的作用.方法 以高脂饮食喂养3周龄的SD雄性大鼠至12周龄,监测正常饮食与高脂饮食两组大鼠的体质量与血压的变化,并以Western blot和HPLC等方法比较两组大鼠的的肾脏各部分20-HETE活性的差异.结果 高脂饮食大鼠体质量和MBP在第12周龄明显较正常饮食组增高(414±7)vs(357±5)g,(132±8)vs(108±5)mm Hg,P<0.05),高脂饮食大鼠无论是全肾,还是肾实质各部分的20-HETE活性均明显下降,同时Western blot表明产生20-HETE的CYP4A表达也出现相应的变化.结论 高脂饮食诱导的幼年型肥胖相关性高血压与肾实质的20-HETE下调有关.

Objective To study renal's 20-hydroxyeicosatetraenoic acid (20-HETE) down-regulation in hypertension induced by high-fat diet. Methods SD male rats were fed with high fat diet from 3-weeks old to 12-weeks old ,compared the weight and MBP changes between high-fat diet and normal diet,analysed 20-HETE activity with RP HPLC and Western blot in different parts of kidney. Results Weight and MBP increased in high fat diet groups at the 12-weeks old(414±7) vs(357±5)g,(132±8)vs(108±5)ram Hg,P <0.05). There were significant down-regulation changes of 20-HETE activities between two groups, no matter in whole kidney or different part of renal parenchyma. And the same changes were found in CYP4A expression with western blot. Conclusion these results demonstrated that juvenile rats with obesity related hypertension induced by high-fat diet were related to the downregulation of 20-HETE in renal parenchyma.
