目的 观察冷冻联合重组人肿瘤坏死因子-α( rhTN F-α)治疗C6大鼠脑胶质瘤治疗的协同作用.方法 以原位末端转移酶标记(TUNEL)法检测肿瘤细胞的凋亡,采用免疫组织化学检测增殖细胞核抗原(PCNA)的表达;行核磁共振(MRI)检查,测量肿瘤体积,计算抑瘤率.动态观察各组荷瘤鼠生存期和rhTNF-α对荷瘤鼠的不良反应.结果 联合治疗组肿瘤细胞凋亡、PCNA的表达、肿瘤体积和生存期与其他各组比较差异有统计学意义(P<0.01),且该组对肿瘤生.长的抑制率(60.38%;89.48%)明显大于冷冻组(42.69%;66.31%)和TNF-α组(21.68%;49.01%),且大于理论抑瘤率(82.82%).rhTNF-α对实验动物有不良反应.结论 冷冻和rhTNF-α对C6联用的抗肿瘤作用进一步加强,具有协同作用.

Objective To observe the synergistic effects of freeze combination with recombinant human tumor necrosis factor-α (rhTNF-α) on the treatmemt of rat C6 brain glioma.Methods TdT-medi-ated dUTP nick end labeling (TUNEL) was applied to detect apoptosis.Proliferating cell nuclear antigen (PCNA) expression in tumor cell was examined by SP immunohistochemical staining.Meanwhile,the tumor volume were measured by MRI and tumor growth inhibition rate was acquired.The survival period of rats and the poison side effect of rhTNF-α were dynamically observed.Results After comparing the combined group with the other three groups,the statistical significance was exhibited about PCNA expression,tumor volume and survival period ( P < 0.01 ).The inhibition rate of tumor growth in combined group (60.38%; 89.48% )was significantly higher than in cryotherapy group (42.69%; 66.31% )and in rhTNF-α group (21.68% ; 49.01% ),and even larger than tbeories inhibition rate ( 55.1 1% ; 82.82% ) of tumor growth.At the same time,the certain poison side effect to the experiment animal was caused by rhTNF-α.Conclusion The combination of freeze and rhTNF-α could strengthen efficiency of antitumor and had synergic effects in the treatment of glioma.
